钙羟磷灰石皮下注射联合维生素C口服对裸鼠皮肤胶原蛋白的影响 被引量:2

The Effects of Calcium Hydroxylapatite Subcutaneous Injection Combining Oral Vitamin C Administration on Skin Collagen in Nude Mice

摘要 目的探讨钙羟磷灰石(calcium hydroxylapatite,CaHA)皮下注射联合维生素C口服对裸鼠皮肤胶原蛋白合成的影响。方法 80只BALB/c裸鼠随机分成对照组、CaHA组(CaHA皮下注射组)、VitC组(VitC口服组)、CaHA+VitC组(CaHA皮下注射联合VitC口服组)。分别于第6周和第12周观察裸鼠背部注射部位皮肤外观,第12周HE和Masson染色观察各组胶原蛋白的含量,组织影像显微成像分析系统对各组胶原蛋白含量定量分析,检测血清谷草转氨酶、谷丙转氨酶、肌酐、血糖、总胆固醇及甘油三酯水平,并进行统计学分析。结果第6周与第12周各组裸鼠注射CaHA部位皮肤外观无红肿、感染及坏死等现象。第12周Masson染色显示,CaHA+VitC组胶原密度明显高于其他组(P<0.05)。其中CaHA+VitC组与对照组差异(P<0.01),CaHA+VitC组与VitC组差异(P<0.01),CaHA+VitC组与CaHA组差异(P<0.05)均有统计学意义。各组血清谷草转氨酶、谷丙转氨酶、肌酐、血糖差异无统计学意义(P>0.05),CaHA+VitC组及VitC组血清总甘油三酯较对照组有显著降低(P<0.05),VitC组血清总胆固醇较对照组有显著降低(P<0.05),差异均有统计学意义。结论 CaHA皮下注射联合维生素C口服可增加裸鼠皮肤胶原蛋白合成,且对血清主要生化指标无不良影响。 Objective To investigate the effects of calcium hydroxylapatite(CaHA)subcutaneous injection combining oral Vitamin C administration on skin collagen in nude mice.Methods The 80 nude mice were randomly and equally divided into 4 groups:control group receiving neither CaHA subcutaneous injection nor oral vitamin C administration,CaHA group receiving CaHA subcutaneous injection only,VitC group receiving oral vitamin C administration only,and CaHA+VitC group receiving both CaHA subcutaneous injection and oral vitamin C administration. At 6 and 12 week,the skin appearance of the injected site was observed. After 12 weeks,all the nude mice were sacrificed and skin tissue specimens were resected from the back of nude mice followed by histopathologic examination and Masson staining for the detection of collagen changes. Serum glutamic oxaloacetic transaminase,glutamic-pyruvic transaminase,creatinine,glucose,total cholesterol and triglyceride were detected as well. Statistical analysis was performed.Results At 6 and 12 week,no erythema,swelling,infection and necrosis response were observed at the injected skin. At 12 week,Masson staining of CaHA+VitC group showed higher density of collagen in dermis by comparison with the other three groups(all P〈0.05). Collagen density was statistically higher in CaHA+VitC group than in the control group(P〈0.01),in CaHA+VitC group than in VitC group(P〈0.01),and in CaHA+VitC group than in CaHA group(P〈0.05). No significant difference was observed in serum glutamic oxaloacetic transaminase,glutamic-pyruvic transaminase,creatinine and glucose in 4 groups(P〉0.05). Serum triglyceride was statistically lower in CaHA+VitC group and VitC group than in control group(P〈0.05),and total cholesterol was statistically lower in VitC group than in control group(P〈0.05).Conclusion CaHA subcutaneous injection combining oral Vitamin C administration increases skin collagen synthesis in nude mice and has no adverse effect on serum glutamic oxaloacetic transaminase,glutamic-pyruvic transaminase,creatinine,glucose,total cholesterol and triglyceride.
