摘要
目的研究肺癌mdig基因对组蛋白H3及H4部分位点赖氨酸残基甲基化状态的作用,为肺癌的诊断和治疗建立基础。方法 随机选择2010年12月-2015年12月我院收治的肺癌患者200例(病例阶段为T1-T4期)作为本次研究的对象,对患者癌组织和癌旁组织中UTX与Rb的mRNA和蛋白质含量进行检测,同时检测肺癌及癌旁组织中H3K27me3和Rb蛋白含量以及UTX去甲基化酶活性,以确定UTX表达及功能异常在肺癌发生、发展和预后中的作用。结果 肺癌患者不同病理期的癌组织和癌旁组织中UTX与Rb的mRNA和蛋白质含量不同,肺癌患者肺癌组织及癌旁组织中H3K27me3和Rb蛋白含量以及UTX去甲基化酶活性差异有统计学意义(P〈0.05),对肺癌mdig基因肺癌细胞中的H3K27me3含量进行检验,沉默mdig肺癌细胞、过表达mdig肺癌细胞中H3K27me3含量无明显变化(P〉0.05)。结论 组蛋白去甲基化酶、UTX、Rb,在肺癌组织的发生和发展中发挥着重要的作用,其可以抑制肿瘤细胞的发展,控制癌细胞的扩散,加强对组蛋白H3及H4部分位点赖氨酸残基甲基化状态的检测和研究,对肺癌的治疗和诊断有重要的意义。
Objective To research the effect of lung cancer mdig gene (mineral dust - induced gene) on methylation status of lysine residues on h cancer.Methods 200 patients istone H3 and H4 partial loci,and to provide basis for diagnosis and treatment of lung with lung cancer (T1-T4) treated in our hospital from December 2010 to December 2015 were randomly selected,and UTX,Rb mRNA and protein content of cancer tissue and para-carcinoma tissue were detected,as well as the H3K27me3 and Rb protein content,and activity of UTX demethylase of cancer tissue and para- carcinoma tissue,to determine the effect of UTX expression and dysfunction in development,progress,and outcome of lung cancer.Results The UTX and Rb of mRNA and protein content of cancer tissue and para-carcinoma tissue were various among patients with lung cancer at different stages,and there were significant differences in H3K27me3 and RB protein content,and activity of UTX demethylase of cancer tissue and para-carcinoma tissue in lung cancer patients (P〈 0.05).According to the result of H3K27me3 content detection in lung cancer mdig gene lung carcinoma cells,there was no significant difference in H3K27me3 content between silent mdig lung carcinoma cells and over-expressed mdig lung carcinoma cells (P〉0.05).Conclusion Histone demethylase,UTX,and Rb play an important role in development and progress of lung cancer tissues by inhibiting development of cancer cells and controlling spread of cancer cells.More studies on methylation status of lysine residues on histone H3 and H4 partial loci have significant meanings for treatmerit and diagnosis of lung cancer .
