摘要
目的 建立壮药了刁竹全草定性鉴别方法和含量测定方法,为了刁竹的质量控制提供参考。方法 采用性状、显微、薄层层析(TLC)鉴别方法和高效液相色谱(HPLC)含量测定方法,对了刁竹进行定性和定量检测,其中HPLC色谱柱为Agilent ZORBAX SB-C18柱,流动相为甲醇-水(55 ∶45),检测波长为274 nm,流速1.0 ml/min,柱温30℃。结果 在TLC鉴别中,两种鉴别方法均可见特征斑点,具有专属性,且特征性强、重复性好。应用HPLC法,当丹皮酚进样量为0.004328~0.2164 μg时,进样量与峰面积呈良好的线性关系:Y=5053.4X-1.7687,r=0.9998,平均加样回收率为99.42%;10批药材丹皮酚含量为0.15%~1.16%,平均为0.59%。结论 建立的了刁竹全草定性鉴定和定量测量标准可有效控制该药材的质量。
Objective To establish the methods of identification and content measurement for Zhuang medicine Cynanchura pan/cu/atum,thus to provide the reference for quality control of Cynanchum paniculatum. Methods Qualitative and quantitative measurement for Cynanchum paniculatum were conducted using macroscopical, microscopic identification, thin-layer chromatography(TLC) and high performance liquid chromatography(HPLC) method. Agilent ZORBAX SB-C18 column was used as chromatographic column, and methanol-water(55 : 45 ) as mobile phase in HPLC. The detection wavelength was 274 nm, the flow velocity was 1.0 ml/min, and temperature was kept at 30℃ in HPLC. Results In identification with TLC,characteristic spot was observed in the two identification methods. And the methods obtained specificity,obvious characteristics and good repeatability. HPLC method was used, when the sample size of Cynanchum paniculatum was between O. 004328 μg and 0.2164 μg, a good linear relationship was found between the sample size and peak area:Y = 5053.4X - 1. 7687, r = 0. 9998, and the average percent recovery of sample was 99.42%. Of ten batches of Cynanchum paniculatum,the contents of paeonol were 0.15% - 1.16% ,and the average content was 0. 59%. Conclusion The established standards of qualitative identification and quantitative measurement can effectively control the quality of Cynanchum paniculatum.
