亚洲带绦虫实验感染对乳猪肝细胞生长抑制率和MAPK通路主要激酶基因表达的影响 被引量:2

摘要 本研究旨在探讨亚洲带绦虫实验感染乳猪后不同时间囊尾蚴寄生处肝组织细胞生长抑制率和MAPK通路主要激酶基因表达的变化。将采自贵州省都匀市的亚洲带绦虫孕节解剖后,用生理盐水反复清洗、离心沉淀收集虫卵。20日龄约克二元施格杂交乳猪29头,随机分为实验组15头和对照组14头,实验组以定量虫卵15万个/头灌胃感染。于感染后第15、45和75d处死乳猪,取实验组囊尾蚴寄生处肝组织和对照组肝组织,采用cck-8法检测各组肝细胞的生长抑制率,并用实时荧光定量PCR检测肝细胞ERK1、JNK1、p38基因mRNA的相对表达量。结果显示,感染后第15、45和75d,cck-8法测定实验组肝细胞的生长抑制率分别为(23.11±1.26)%、(25.15±1.26)%和(20.72±1.3)%。实时荧光定量PCR检测实验组乳猪肝细胞的ERK1和p38mRNA表达水平均较对照组上调,而JNK1mRNA表达水平均较对照组下调,随感染时间延长,ERK1和p38mRNA表达水平均明显下调。结果表明亚洲带绦虫囊尾蚴感染可明显影响乳猪肝细胞ERK1和p38基因mRNA水平。 The aim of present study is to investigate the growth inhibition rate of hepatic cells and the expressions of MAPK- related kinase genes in liver cells of the porkets experimentally infected with Taenia asiatica. The adult worm of T. asiatica was collected and identified from taeniasis patients in Duyun C. , Guizhou Province. The eggs were collected from gravid proglottids and prepared by repeated washing and centrifugation. Twenty-nine hybrid porkers aged in 20 days were randomly assigned into experimental group (n = 15) and control group (n = 14). Porkets in experimental group were infected with 1.5 x 105 eggs/pig by stomach infusion. The porkets were sacrificed at the 15th, 45th and 75th day after infection, and liver samples were collected for further experiments. The growth inhibitory rate of hepatic cells was estimated with CCK-8 method, qRT-PCR was employed to detect the expression levels of mRNA of ERK1, JNK1 and p38 genes in hepatic cells of both experimental and control group. CCK-8 test showed that the growth inhibition rates of hepatic cells in experimental group were (23.11 + 1.26)% , (25.15 _+ 1.26)% and (20.72 -+ 1.3)% , respectively, at the 15th, 45th and 75th day after infection, implying statistically significant difference, qRT-PCR detection revealed that the mRNA levels of ERK1 and p38 were up-regulated, while of JNK1 were down-regulated in experimental group than in control group. However, the mRNA levels of ERK1 and p38 in the experimental group decreased obviously along with the extended time of infection. The results indicated that the infection of T. asiatica cystieereus may have an obvious effect on the mRNA levels of ERK1 and p38 in liver cells of porkets.
