摘要 本研究对云南省分离的一株NamDinh病毒(NDiV)进行鉴定,并对其全基因组特征进行分析。采用白纹伊蚊C6/36细胞对2009年在云南省采集的蚊虫进行病毒分离;采用RT—PCR方法对阳性病毒分离物进行鉴定,在电镜下观察其形态特征;采用Mega5软件对病毒全基因组序列进行系统进化分析。结果从一组三带喙库蚊标本中分离出一株NDiV。该病毒为球形颗粒,直径约100~160nm;基因组全长为20126bp,包括6个开放读码框架,与越南NDiV02VN178株和中国深圳SZ11706Z株核苷酸和氨基酸同源性分别为94.0%~100%和97.4%~100%,同属于套式病毒目Mesoniviridae病毒科。本研究获得的NDiV毒株及其全基因组序列特征系云南省首次报告。 A new strain of Nam Dinh virus (NDiV) , an insect nidovirus, isolated in Yunnan province has been identified and its genomewide character was analyzed. Aedes albopictus C5/36 cells were used to isolate virus from mosquitoes collected in Yunnan Province in 2009. The culture supernatant with positive cytopathic effect (CPE) was harvested for virus identification by RT-PCR, and the morphology was observed under transmission electron microscope. Pbylogenetic analysis of virus genomewide sequence was carried out using the Mega 5 program. One isolate was obtained from a Culex tritaeniorhynchus pool and identified as NDiV. The viruses were small spherical particles of approximately 100 - 160 nm. The full- length genome of the new NDiV was 20 126 bp and contained 5 open reading frames (ORFs). With 98. 0% sequence identity, the genome of this new strain was mostly related to the genome of NDiV 02VN178 isolated from Culex mosquitoes in Vietnam. The different ORFs of the two strains showed 94. 0% - 100% sequence identity and 97. 4% - 100% amino acid sequence identity. Phylogenetic analysis showed that the new strain, along with NDiV 02VN178 and another Chinese NDiV strain, was classified into the family Mesoniviridae. This is the first report of NDiV and its genomewide character isolated from Cx. tritaeniorhynchus in Yunnan Province, and may add important information to distribution of the virus.
