腺苷脱氨酶1在肺腺癌组织的表达及其与预后的关系 被引量:1

Expression of adenosine deaminase acting on RNA 1 in lung adenocarcinoma and its correlation with prognosis

摘要 目的探讨腺苷脱氨酶l(ADAR1)在肺腺癌中的表达及其临床意义。方法应用免疫组织化学染色检测48例肺腺癌患者肿瘤组织与癌旁正常组织中ADAR1蛋白表达水平。应用实时定量反转录聚合酶链反应(RT—qPCR)检测肺腺癌患者肿瘤组织与癌旁正常组织中ADAR1基因表达水平。结果肺腺癌组织中ADAR1蛋白阳性表达率显著高于癌旁正常组织(60.4%比20.8%,P〈0.05)。肺腺癌组织中ADAR1基因表达水平显著高于癌旁正常组织(1.58比1.19,P〈0.05)。肺腺癌组织中ADAR1蛋白阳性表达率与肿瘤TNM分期(X2=7.779,P〈0.05)和淋巴结转移(X2=4.516,P〈0.05)密切相关。Kaplan—Meier生存分析显示,ADAR1高表达组患者的总生存期(OS)较低表达组明显缩短,其中ADAR1高表达组(29例)中位生存时间为27个月;低表达组(19例)中位生存时间为56个月,差异有统计学意义(X2=8.916,P〈0.01)。Cox比例风险回归模型分析结果显示,ADAR1蛋白表达水平、TNM分期是影响预后的独立因素(P〈0.05)。结论ADAR1在肺腺癌中高表达与患者预后差密切相关。 Objective To investigate the expression of adenosine deaminase acting on RNA 1 (ADAR1) and its correlation with clinicopathological characteristics and prognosis in lung adenocarcino- ma. Methods The expression of ADAR1 protein in lung adenocarcinoma tissues ( n = 48) and adjacent normal lung tissues (n = 48) was detected by immunohistochemistry. Real - time quantitative polymerase chain reaction ( RT - qPCR) was applied to detect the expression of ADAR1 mRNA in fresh lung adenocar- cinoma tissues and adjacent normal lung tissues. Correlation of the expression of ADAR1 with clinicopatho- logical characteristics and overall survival of lung adenocarcinoma patients was analyzed. Results The ex- pression of ADAR1 protein was significantly higher in lung adenocarcinoma tissues than in adjacent normal lung tissues (60. 4% vs. 20. 8%, P 〈 0. 05). The expression of ADAR1 mRNA was significantly higher in lung adenocarcinoma tissues than in adjacent normal lung tissues (1.58 vs. 1.19, P 〈 0. 05). The ex- pression of ADAR1 protein was significantly correlated with TNM stage (X2 = 7. 779, P 〈 0. 05) and lymph node metastasis (X2 =4. 516, P 〈0. 05). The median overall survival time was longer in the low ADAR1 expression group than in the high expression group (56 months vs. 27 months, P 〈 0. 05) by Kaplan - Meier survival analysis. The cox multivariate analysis showed that ADAR1 expression and TNM stage were independent prognostic factors (P 〈 0. 05). Conclusion ADAR1 is overexpressed in lung adenocarcino- ma. High expression of ADAR1 correlates with poor survival in lung adenocarcinoma.
