摘要
目的观察Kiss-1基因通过下调促细胞丝裂原细胞外激酶-胞外信号调节激酶.丝裂原活化蛋白激酶信号传导通路(MEK—ERK—MAPK)途径对人结直肠癌HCTl16细胞侵袭、转移能力的影响。方法重组LV.Kiss-1基因慢病毒为载体转染人结直肠癌HCT116细胞,实验分为空白对照组(CON组)、慢病毒空载体阴性对照组(NC组)、Kiss-1基因过表达组(OE组)。细胞计数试剂盒(CCK-8)法检测转染前后细胞增殖能力的变化,Transwell法分别检测转染前后细胞的侵袭和迁移能力的变化。应厢Westernblot法检测3组细胞中MAPK信号通路的关键分子丝裂原细胞外激酶(MEK)表达含量的变化以及下游效应蛋白肌球蛋白轻链(MLC)磷酸化水平的变化。结果OE组HCTll6细胞中MEK的表达含量为0.3920±0.0107,较CON组的0.8284±0.0167、NC组的0.8405±0.0092明显降低(P〈0.05),下游效应蛋白MLC磷酸化水平OE组为0.1783±0.0072,较CON组的0.5246±0.0122、NC组的0.5441-4-0.0135亦明显下降(P〈0.05),差异有统计学意义。OE组较CON、NC组细胞增殖受到明显抑制(P〈0.05),细胞侵袭、迁移能力亦出现明显抑制(P〈0.05)。结论LV-Kiss-1基因慢病毒转染人结直肠癌HCTl16细胞后,可能通过MEK—ERK—MAPK信号传导通路途径抑制其增殖、侵袭和迁移能力。
Objective To study the inhibitor effect of Kiss - l gene on invasion and metastasis abil- ity of HCTll6 human colorectal carcinoma cells via down- regulating mitogen extraeellular kinase (MEK)/ extraeellular signal regulated kinase (ERK) and mitogen activated protein kinase (MAPK) signal transdue- tion pathway. Methods The lentiviral vectors of Kiss - 1 gene were constructed and transfeeted into HCTll6 cells. Primary HCTII6 human colorectal cancer cells, non -carrier transfected cells and lentiviral vector transfected cells were divided respectively into: blank control group ( CON group ) , non - carrier control group (NC group) and over-expression group (OE group). The cell counting kit -8 (CCK-8) and Tran- swell chamber methods were used to detect the changes of cell proliferation, invasion and migration ability before and after transfeetion. Western blotting was used to detect the expression of inhibitive protein of MEK - ERK - MAPK signal pathway and the expression of downstream effeetor ( MLC ) before and after transfection. Results The MEK expression level in QE group (0. 392 0 ± 0. 010 7) decreased significantly compared to that in CON group (0. 828 4 ±0. 016 7) and NC group (0. 840 5 ±0. 009 2,P 〈0. 05). The MLC expression level in QE group (0. 178 3 ± 0. 007 2) also decreased significantly compared to that in CON group (0.524 6±_0.012 2) and NC group (0.544 1 ±0.013 5,P〈0.05). Compared to that in CON and NC group, cell proliferation, invasion and migration ability in OE group was inhibited obviously with statistically significant difference (P 〈 0. 05 ). Conclusion After lentiviral transfection, Kiss - 1 gene may inhibit cell proliferation, invasion and migration ability of HCT116 human colorectal carcinoma ceils via MEK - EI±K - lql)tPK s!gnal pathway.
