甘氨酸甲基转移酶在胃癌与癌旁组织中的表达及临床意义

Expression and clinical significance of glycine N - methyltransferase in gastric cancer and para - carcinoma tissues

目的探讨胃癌与癌旁组织中甘氨酸甲基转移酶（GNMT）的表达及其临床意义。方法选取我院94例胃癌患者，分别检测这些患者胃癌与癌旁组织中GNMT的表达水平与胃癌大小、TNM分期及分化程度的关系。同时分别运用实时定量反转录聚合酶链反应（RT—qPCR）和Westernblot检测癌旁与癌组织内GNMTmRNA及蛋白的表达；利用酶联免疫吸附试验（ELISA）法检测癌组织与癌旁组织中GNMT的含量。结果94例癌旁组织中GNMTmRNA及蛋白的表达量明显高于癌组织（P〈0．05）。ELISA检测得出胃癌肿块≥4．0cm者GNMT表达量为（566．71±233．65）ng／ml，胃癌肿块〈4．0cm者GNMT表达量为（764．60±187．38）ng/ml。分化程度越高的癌组织内GNMT表达量越高，TNM分期越高的肿瘤组织内GNMT的表达量越低。结论GNMT在胃癌的发生和发展过程中发挥抑制癌组织生长的作用。

Objective To analyze the expression and clinical significance of glycine N - methyl- transferase （GNMT） in gastric cancer and para - carcinoma tissues. Methods Ninety - four eases of gas- tric cancer were selected from January 2013 to January 2016. The expression of GNMT in gastric cancer and para -carcinoma tissues was detected, and the relationship between the GNMT expression and tumor size, TNM stage and degree of tumor differentiation was analyzed. Meanwhile, the real - time quantitative reverse transeriptase - polymerase chain reaction （ RT - qPCR） and Western blotting were applied to exam- ine the GNMT mRNA and protein expression in cancer and para - carcinoma tissues. The enzyme linked immunosorbent assay （ELISA） was used to assay the expression of GNMT in cancer and para - carcinoma tissues. Results The GNMT mRNA and protein expression was higher in para - carcinoma tissues than in cancer tissues （P 〈 0. 05 ）. The expression of GNMT was lower in cancer tissues with tumor diameter over 4 cm [（ 566.71 ± 233.65 ） ng/ml ] than in those with cancer tissues less 4 cm [（ 764.60 ± 187. 38） ng/ml]. The GNMT expression was higher in the well differentiated tumor tissues than in poorly differentiated tissues. Conclusion GNMT could be used as a cancer suppressor gene in the occurrence and development of gastric cancer.
