摘要
目的:建立了同时测定马黛茶生物碱(可可碱和咖啡因)含量的高效液相色谱法。方法:采用华谱Unitary C_(18)(150 mm×4.6 mm,5μm)色谱柱,以甲醇和0.5%甲酸水溶液为流动相,流速为1.0 m L·min^(-1),检测波长为280 nm。结果:马黛茶中可可碱和咖啡因在5.010~200.0μg·m L^(-1)呈现良好线性关系,r均大于0.999,RSD均小于1.70%,平均回收率分别为96.59%和97.20%。结论:该方法简便、准确、灵敏、重复性好,可用于马黛茶的质量控制。
Objective : An HPLC method was established for simultaneous determination of theobromine and caffeine-- two bioactive compounds from Mate Tea (llex paraguariensis). Methods : An Unitary C18 ( 150 mm ×4.6mm,5μm) column was used with the mobile phase of methanol and water containing 0. 5% of formic acid at the detection wave length of 280 nm and the flow rate of 1.0 mL·min^-1. Results : The two alkaloids were successfully separated in 30min, and the calibration curves of theobromine and caffeine were linear in the range of 5. 010-200. 0 μg·mL^-1 with correlation coefficient of 0. 999. The recovery rates were 96. 59% and 97.20%, respectively with RSD less than 1.70%. Conclusion: The established method is simple, accurate, sensitive and good repeatability, which can be applied for the quality control of Mate Tea ( I. paraguariensis ).
