Optimization of Extraction Process of Volatile Oil from Ramulus Cinnamomi and Atractylodes Rhizome from Tingli Shengmai Decoction by Multi-index Orthogonal Test

摘要 目的:优选葶苈生脉方中桂枝-白术挥发油的最佳提取工艺。方法:采用水蒸气蒸馏法提取桂枝、白术挥发油,并用高效液相色谱法对桂皮醛、白术内酯I进行含量测定,以收油率、桂皮醛、白术内酯I含量的综合评分为指标,通过L9(3^4)正交试验,考察加水量(倍)、浸泡时间(h)、提取时间(h)三个因素对桂枝、白术挥发油提取工艺的影响。结果:桂枝、白术挥发油提取的最佳工艺为10倍量的水,浸泡6h,提取4h。结论:正交试验法优选的桂枝、白术挥发油的提取工艺合理可靠,可为葶苈生脉方的开发提供依据。 Objective: To optimize the extraction technology of volatile oil from Atractylodes Rhizome and Ramulus Cinnamomi. Methods: Ramulus Cinnamomi and Atractylodes Rhizome volatile oil was extracted by steam distillation. Contents of atractylenolide I and einnamaldehyde were determined by HPLC. Orthogonal test was adopted to optimize extraction process with composite score of content of volatile oil of Atractylodes Rhizome, cinnamic aldehyde and atractylenolide as index, the amount of water, soaking time and extraction time as factors. Results: Ramulus Cinnamomi and Atractylodes Rhizome volatile oil extraction optimum process is: adding 10 times amount of water, soak 6 hours, extraction for 4 hours. Conclusion: The optimum extraction process of water vapor distillation method is reasonable and reliable for industrial production of Tingli Shengmai decoction.
