CyclinD1与CyclinD3在非小细胞肺癌中的表达及意义 被引量:8

Expressions of CyclinD1 and CyclinD3 in Non-small Cell Lung Cancer Patients and Their Significance

摘要 [目的]探讨细胞周期蛋白D1(Cyclin D1)与细胞周期蛋白D3(Cyclin D3)在非小细胞肺癌(NSCLC)中的表达及临床意义。[方法]免疫组织化学法检测Cyclin D1、Cyclin D3蛋白在271例NSCLC及相应正常组织中的表达情况,分析其相关性及其与临床特征的关系。[结果 ]Cyclin D1和Cyclin D3定位于细胞质和细胞核中,癌组织与正常组织中阳性表达率差异显著(P均为0.000),同一患者癌组织中Cyclin D3表达高于Cyclin D1。Cyclin D1、Cyclin D3蛋白表达在不同性别、年龄、吸烟史、肿瘤大小及位置、病理分型组中均无统计学差异(P均>0.05)。在不同病理分级、临床TNM分期及淋巴结是否转移组中,Cyclin D1表达均无统计学差异(P>0.05),而Cyclin D3表达差异均显著(P<0.05)。在总NSCLC、腺癌、鳞癌中,Cyclin D1和Cyclin D3表达的相关性检验显示两者均具有正相关性,r分别为0.332、0.377、0.308(P均为0.000)。Cyclin D1低表达患者和高表达患者的中位生存时间接近,无统计学差异(P=0.264);Cyclin D3低表达患者的中位生存时间较高表达者明显延长,差异有统计学意义(P=0.001)。[结论 ]Cyclin D1和Cyclin D3表达可能与NSCLC发生相关,但在NSCLC发展与转移等恶性生物学行为中,Cyclin D3起重要作用,并可作为评估NSCLC恶性生物学行为及预后的参考指标。 [Objective] To investigate the expressions of Cyclin D1 and Cyclin D3 in non-small cell lung cancer(NSCLC) and their significance.[Methods] The expressions of Cyclin D3 and Cyclin D1 in 271 cases with NSCLC and corresponding normal tissues were detected by immunohistochemical method,and their correlation with clinicopathological features was analyzed.[Results] Positive staining of Cyclin D1 and Cyclin D3 was observed in cytoplasm and nucleus,and the expression of Cyclin D3 was higher than that of Cyclin D1 in the same case.The positive expression rates of the Cyclin D1 and Cyclin D3 proteins in NSCLC were significantly higher than those in the corresponding normal tissue(P=0.000).The expressions of Cyclin D1 and Cyclin D3 were not significantly correlated with gender,age,smoking history,tumor size,location and pathological classification(P0.05).There were no significant differences in Cyclin D1 expression(P=0.102,P=0.987,P=0.827),but were significant differences in Cyclin D3 expression(P=0.001,P=0.002,P=0.011) among NSCLCs with different pathological grades,clinical stages and lymph node metastasis.The protein expression levels of Cyclin D1 were positively correlated with those of Cyclin D3 in NSCLC,adenocarcinoma and squamous cell carcinoma(r=0.332,r=0.377,r=0.308;all P=0.000).The median survival time of patients with low expression of Cyclin D1 was not different to patients with high expression level(P=0.2640).The patients with higher expression Cyclin D3 had shorter survival time compared with those with lower expression patients(P=0.0005).[Conclusion] The expression of Cyclin D1 and Cyclin D3 might be associated with the occurrence,development and metastasis of NSCLC,which can be used to evaluate the malignant behavior and prognosis of NSCLC.
