二甲丸对大鼠增生型子宫内膜中ER、PR及BFGF、VEGF的影响 被引量:4

Effect of Erjia pills on estrogen receptor,progesterone receptor,basic fibroblast growth factor,and vascular endothelial growth factor in hyperplastic endometrium in rats

摘要 目的:观察益肾软坚之二甲丸对大鼠增生型子宫内膜中雌激素受体(ER)、孕激素受体(PR)以及碱性成纤维细胞生长因子(BFGF)、血管内皮生长因子(VEGF)表达的影响,探讨其治疗功能性子宫出血之子宫内膜增生的作用机制。方法:将健康SD大鼠80只随机分为空白组、模型组和二甲丸高、中、低剂量组及西药组,其中模型组和西药组各14只,其余每组各13只。空白组常规饲养,其余组均行去势术,术后除空白组外均给予补佳乐灌胃建立大鼠子宫内膜增生模型。空白组与模型组仅予等量蒸馏水灌胃,二甲丸高、中、低剂量组分别给予相应剂量二甲丸汤剂灌胃,西药组给予地屈孕酮研磨灌胃,均连续灌胃14d。采用免疫组化SP法检测各组大鼠子宫内膜中ER、PR、BFGF、VEGF的表达水平。结果:通过干预后二甲丸各组BFGF、VEGF均明显下降;相关分析发现,治疗前后比较BFGF、VEGF与ER呈显著正相关,而与PR则呈显著负相关,这种变化与正常大鼠子宫内膜相符合。结论:益肾软坚法可能通过ER、PR的介导作用,降低BFGF、VEGF的表达,进而抑制血管过度增殖,从而达到阻断子宫内膜异常增生的目的。 Objective: To investigate the effect of Erjia pills for nourishing kidney and softening hard mass on the expression of estrogen receptor( ER),progesterone receptor( PR),basic fibroblast growth factor( b FGF),and vascular endothelial growth factor( VEGF),as well as its mechanism of action in the treatment of endometrial hyperplasia in functional uterine bleeding. Methods: A total of 80 healthy Sprague- Dawley rats were randomly divided into blank group,model group,high-,medium-,and low- dose Erjia pill groups,and Western medicine group,with14 rats in the model group and the Western medicine group and 13 rats in the other groups. The rats in the blank group were given routine feeding,and those in the other groups were ovariectomized and given Progynova by gavage to establish the rat model of endometrial hyperplasia. The rats in the blank group and the model group were given distilled water of the same volume by gavage,those in the high-,medium-,and low- dose Erjia pill groups were given Erjia decoction by gavage,and those in the Western medicine group were given ground dydrogesterone by gavage. All the rats were treated by gavage for 14 days. Immunohistochemistry substance P was used to measure the expression of ER,PR,b FGF,and VEGF in the endometrium in rats. Results: After intervention,the Erjia pill groups showed significant reductions in b FGF and VEGF. Correlation analysis showed that b FGF and VEGF were significantly positively correlated with ER but negatively correlated with PR. Such changes were consistent with those in the endometrium in normal rats. Conclusion: The method of nourishing kidney and softening hard mass can reduce the expression of b FGF and VEGF through mediating ER and PR,inhibit excessive vascular proliferation,and thus block abnormal endometrial hyperplasia.
