摘要
目的:建立野菊花注射液中主要药效成分物质绿原酸、木犀草素-7-O-β-D-葡萄糖苷和蒙花苷的含量测定方法。方法:采用RP-HPLC方法,色谱柱为Ultimate^(TM) XB-C18色谱柱(250mm×4.6mm,5μm),采用0.6%冰醋酸水溶液-甲醇为流动相进行梯度洗脱,流速为1.0m L·min^(-1),检测波长为327nm,柱温为30℃。结果:绿原酸、木犀草素-7-O-β-D-葡萄糖苷和蒙花苷分别在5.92~142.1μg/m L、1.04~24.96μg/m L、4.32~103.7μg/m L与峰面积呈良好的线性关系;平均回收率和RSD分别为98.7%和1.9%、99.0%和1.8%、99.1%和2.2%。结论:所建立的HPLC法准确、快速、灵敏度高,可用于野菊花注射液中主要药效成分物质绿原酸、木犀草素-7-O-β-D-葡萄糖苷和蒙花苷的含量测定,可以用于野菊花注射液质量控制的提高。
Objective: To investigate the method for the determination of major effective constituents,chlorogenic acid,luteolin- 7- O- β- D- glucoside,and linarin,in Yejuhua injection. Methods: Reverse- phase high- performance liquid chromatography was performed on an Ultimate^(TM) XB- C18column( 250 mm × 4. 6mm,5μm) with a mobile phase of 0. 6% glacial acetic acid solution- methanol for gradient elution at a wavelength of 327 nm,a flow rate of 1. 0 ml / min,and a column temperature of 30°C. Results: Chlorogenic acid,luteolin- 7- O- β- D- glucoside,and linarin showed a good linear relationship between concentration and peak area within the ranges of 5. 92-142. 1 μg / m L,1. 04 ~ 24. 96 μg / m L,and 4. 32- 103. 7 μg / m L,respectively,and the average recovery rates were98. 7%( RSD = 1. 9%),99. 0%( RSD = 1. 8%),and 99. 1%( RSD = 2. 2%),respectively. Conclusion: The HPLC method established is accurate,time- saving,and highly sensitive and can be used for the determination of chlorogenic acid,luteolin- 7- O- β- D- glucoside,and linarin in Yejuhua injection,as well as the quality control of Yejuhua injection.
