广西良种杉木组培育苗快繁技术体系的优化 被引量:8

摘要 以广西良种杉木优良无性系—柳327幼林萌芽条为外植体,通过采用正交设计方法对培养基激素组合、培养室光照强度和温度等杉木快繁条件进行优化。结果表明:继代丛芽转接时,丛芽中高≥1.3cm的单芽全部切除,接种于改良1/2 MS+6-BA 0.7mg/L+KT 0.3mg/L+IBA 0.1mg/L培养基内,培养35d,芽增殖系数达5.2,有效芽38株/瓶(500mL三角瓶);将高≥1.3cm的组培单芽扦插于改良1/2 MS+KH2PO4100 mg/L+IAA 3mg/L+IBA 2mg/L+ABT60.8mg/L培养基内,然后置于温度20~25℃,光照2500~6000Lx的培养环境条件下,培养45d,平均生根率达98.3%,根系数量为5.6条/株。经过优化的杉木组培育苗体系达到了产业化生产的需求,为广西良种杉木的快速繁育提供了技术支撑。 Using the clone of improved varieties of Cunninghamia lanceolate, liu327 , as the explant, several conditions such a hormone combination, light intensity, and temperature were optimized based on the method of orthogonal design in Guangxi in this study. The results showed that after subculture of 35 days, the proliferation of shoots was 5. 2 fold and the number of effective buds was 38 plants per flask if single buds higher than 1. 3 cm were cut off from clustered shoots and transferred to the modified 1/2 MS medium + 0. 7 mg/L 6-BA + 0. 3 mg/L KT +0. 1 mg/L IBA during the subculture of shoots. After rooting culture of 45 days with 20~25 ℃ temperature and 2 500~6 000Lx light intensity, the average of rooting percentage was 98. 3%, each rooted plantlet has 5. 6 roots and robust if single buds higher than 1. 3 cm were cultured in the rooting medium with the modified 1/2 MS medium+ 100 mg/L KH2 PO4+ 3 mg/L IAA + 2 mg/L IBA + 0. 8 mg/L ABT. This achieved the requirement for the com-mercial production of tissue cultured seedlings in Cunninghamia lanceolata and provided technical support for the rapid development of improved varieties of Cunninghamia lanceolata in Guangxi.
