原子吸收光谱法测定血清中不同化学形态的钴与镍 被引量:1

摘要 采用乙醇低温(4℃)沉淀蛋白法,将血清中的钴、镍分为结合态和非结合态,利用石墨炉原子吸收光谱法灵敏度高、取样量少的优点,测定血清中的钴、镍2种元素的总量及非结合态含量,通过差减法求出血清中结合态元素含量,从而建立钴、镍2种元素化学形态的分离分析方法。以胎牛血清为例,详细研究了仪器工作条件的优化、沉淀剂种类及用量的选择、测定方法准确度的考察和测定方法的精密度及检出限。结果表明,该方法钴、镍的相对标准偏差分别为4.5%和2.5%,加标回收率分别为84%~98%和91%~94%,检出限分别为6.36,8.60μg/L,测定结果可靠。 Separate cobalt and nickel in the serum into the forms of combination and non-combination by the use of ethanol at low temperature(4 ℃) protein precipitation method. Because the sensitivity of GFAAS is high and the sampling is low,this study uses it to analysis the total amount of Co and Ni in serum and the amount of the forms of non-combination. The amount of the forms of combination of Co and Ni is obtained by calculation,so as to establish two chemical forms of Co and Ni separation element analysis method. Take fetal bovine serum for example,a detailed study are as follow:working conditions for the choice of instruments, the type and amount of precipitation agent of choice, determination of the accuracy of the inspection,determination of precision and seizure the limited. The results show that the cobalt and nickel,respectively,the relative standard deviation of 4.5% and 2.5%,and recovery are 84%~98% and 91%~94%,respectively,the detection limit6.36,8.60 μg/L. The result is accurate and reliable.
