紫外-分光光度法测定A群脑膜炎球菌多糖含量的不确定度评估 被引量:2

摘要 目的对紫外-可见分光光度法测定A群脑膜炎球菌多糖含量的不确定度进行评估。方法根据测量过程,分析评估A群脑膜炎球菌多糖含量测定过程中的不确定度来源,计算各分量的不确定度,建立数学模型,计算出合成标准不确定度uc和扩展不确定度U。结果 A群脑膜炎球菌多糖含量测定的合成标准不确定度uc和扩展不确定度U分别为21和42μg/ml(k=2)。结论磷标准溶液的配制稀释是影响紫外-可见分光光度法测定A群脑膜炎球菌多糖含量不确定度的主要因素,可从此处着手改善方法。 Objective To evaluate the uncertainty of determination of polysaccharide content in Neisseria meningitidis serogroup A polysaccharide by spectrophotometry. Methods Uncertainty sources were analyzed to establish a mathematics model, based on which uncertainty components were calculated to evaluate the combined relative standard uncertainty uc and the expanded uncertainty U. Results The combined relative standard uncertainty ucof polysaccharide content in Neisseria meningitidis serogroup A polysaccharide was 21 μg / ml, while the expanded uncertainty U was 42 μg / ml(k = 2).Conclusion The primary source of uncertainty for the determination of polysaccharide content in Neisseria meningitidis serogroup A polysaccharide by spectrophotometry is the preparation and dilution of standard phosphorus solution, which may be a route to improvement of the method.
