集落刺激因子、干扰素-β及肿瘤坏死因子-α在溃疡性结肠炎中的检测价值 被引量:4

摘要 目的探讨集落刺激因子(CSF)、干扰素-β(IFN-β)及肿瘤坏死因子-α(TNF-α)在溃疡性结肠炎患者血清和结肠黏膜组织中表达情况及检测价值。方法选择符合诊断标准的活动期溃疡性结肠炎患者30例作为患者组,以及同期健康对照者30例作为正常组,分别应用酶联免疫法(ELISA)及荧光定量PCR法检测2组血清及结肠黏膜组织中CSF、IFN-β及TNF-α表达情况。结果患者组血清CSF、TNF-α水平及结肠黏膜组织中CSF mRNA、TNF-αmRNA表达量均明显高于正常组(P均<0.05),血清IFN-β水平及结肠黏膜组织中IFN-βmRNA表达量均明显低于正常组(P均<0.05)。CSF、IFN-β、TNF-α变化与溃疡性结肠炎病情存在相关性。结论溃疡性结肠炎患者血清及结肠黏膜组织中CSF、TNF-α表达水平均明显增高,IFN-β表达水平均明显降低,CSF、IFN-β及TNF-α表达水平可作为判断溃疡性结肠炎病情的重要参考指标。 Objective It is to investigate the clinical significance of colony stimulating factor( CSF),interferon-beta( IFN-β) and tumor necrosis factor( TNF- α) in the occurrence and development of ulcerative colitis by detecting the changes of above indexes in serum and colonic mucosal of ulcerative colitis. Methods 30 cases of patients with ulcerative colitis were selected to the patients group,and 30 cases of healthy people were selected to the healthy control group. The serum and colonic mucosa of patients with ulcerative colitis and healthy control group were collected. The expression of CSF,IFN- β and TNF-α were detected by enzyme-linked immunosorbent assay( ELISA) and Real-time PCR respectively. Results The expression of CSF and TNF- α in serum and the levels of CSF mRNA and TNF- α mRNA in colonic mucosa of the patients group were significantly higher than those of the healthy control group( all P〈0.05). the expression of IFN- β in serum and the level of IFN- β mRNA in colonic mucosa were significantly lower than those of the healthy control group( all P〈0.05). The changes of CSF,IFN- β and TNF- α were significantly correlated with the severity of ulcerative colitis. Conclusion The expression of CSF,IFN- β and TNF in serum can be used as an important reference index for judging the condition of ulcerative colitis.
