摘要
目的:评价检测中枢性性早熟(CPP)女童血清mi R-137的临床意义。方法:收集2012年12月至2014年12月在我院门诊就诊的68例CPP女童及68例健康女童血清;培养HEK293细胞,荧光素酶活性分析验证mi R-137与KISS1结合情况;q RT-PCR检测mi R-137的表达;化学发光法检测血清黄体生成素、泌乳素、卵泡刺激素、促甲状腺素、游离甲状腺素、雌二醇水平;酶联免疫吸附试验检测血清吻素的表达。结果:mi R-137与KISS13′UTR结合;CPP女童血清中mi R-137降低,吻素水平升高,两者呈负相关;mi R-137与骨龄、骨龄年龄差呈负相关;Gn RH激发试验中,mi R-137与LH峰值、峰值/基础LH比值呈负相关。结论:mi R-137可与KISS13′UTR结合,可作为CPP辅助诊断的潜在生物学标志物。
Objective To investigate the clinical significance of testing serum kisspeptin in central precocious puberty(CPP) girls. Methods Sixty eight CPP girls and 68 healthy girls was studied from December2012 to December 2014. HEK293 cells were cultured. Luciferase reporter assay was performed to verify the binding of mi R-137 to the 3′UTR of KISS1. Serum mi R-137 level was levaluated by q RT-PCR. Level of serum luteinizing hormone, prolactin, follicle stimulating hormone, thyrotropin, free thyroxine and estradiol was evaluated by chemi-luminescence immunoassay. The level of serum kisspeptin was detected by ELISA. Results Mi R-137 was confirmed to bind to the 3′ UTR of KISS1. The level of serum mi R-137 was downregulated and kisspeptin was enhanced in CPP girls. The expression of mi R-137 and kisspeptin was negatively correlated.Serum mi R-137 level was negatively related to bone age and bone age advancement. According to the results of Gn RH stimulating test, serum mi R-137 was related to peak LH and peak / basal LH ratio. Conclusions Mi R-137 could bind to the 3′UTR of KISS1. Mi R-137 may be a potential biomarker for CPP assisted diagnosis.
