摘要
目的:研究全反式虾青素对环磷酰胺诱导小鼠睾丸氧化损伤的保护作用。方法:将成年60只小鼠随机分为正常对照组、模型组、全反式虾青素低剂量组、全反式虾青素中剂量组、全反式虾青素高剂量组。模型组和不同剂量全反式虾青素组小鼠按40 mg/kg每日1次进行腹腔注射环磷酰胺,连续1wk,同时全反式虾青素低、中、高剂量组分别按照10 mg/kg、15mg/kg、20 mg/kg每日灌胃1次;正常对照组、模型组灌胃等量的食用油。饲养4wk后处死,观察小鼠附睾精子密度、活力、活动力、畸形率,睾丸匀浆测定丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)。结果:与正常对照组相比,模型组精子密度、活力、活动力、SOD、GSH-Px活性均有所下降(P<0.01),精子畸形率、MDA含量有所上升(P<0.01);而与模型组相比,全反式虾青素各剂量组精子密度、活力、活动力、SOD、GSH-Px活性均有所升高(P<0.01),精子畸形率、MDA含量有所下降(P<0.01)。结论:全反式虾青素可以有效抑制环磷酰胺对小鼠睾丸造成的氧化损伤,并可恢复生精功能。
Objective: It was observed that the protective effects of E- astaxanthin on the damage of mice spermatogenesis induced by cyclophosphamide. Methods: The mice were randomly divided into five groups,including normal control,model and low dosage E- astaxanthin,middle dosage E- astaxanthin and high dosage E-astaxanthin. E- Astaxanthin treatment groups and model group in accordance with 40 mg / kg / d by intraperitoneal injection of cyclophosphamide in one week,while the E- astaxanthin treatment groups were given with different dosages E- astaxanthin( 10 mg / kg,15 mg / kg,20 mg / kg). The normal group and model group were given with oil. The treatment period for all groups was four weeks. After all mice were killed,whose testis homogenate were prepared to measure the level of superoxide dismutase( SOD),glutathione perioxidase( GSH- Px) activities and malondialdehyde( MDA) respectively. Sperm density,sperm motility and sperm abnormality rate were also observed. Results: Compared with normal control,testis SOD、GSH- Px activities,sperm density and sperm motility were decreased( P〈0. 01),perm abnormality rate and MDA content were increased( P〈0.01) in model group. Treatment with different dosage E- astaxanthin could significantly increase testis SOD、GSH- Px activities,sperm density and sperm motility( P〈0. 01),decrease sperm abnormality rate and MDA content( P〈0. 01). Conclusion: E- Astaxanthin can effectively inhibit oxygen damage induced by cyclophosphamide,and improve spermatogenic damage.
