H9N2亚型禽流感病毒新疆油鸡分离株NS基因序列测定及进化生物信息分析

Sequencing and bioinformatics analysising of NS gene of H9N2 subtype avian influenza viruses isolated from Xinjiang soy sauce chicken

摘要 为了明确新疆油鸡H9N2亚型禽流感病毒(AIV)非结构蛋白基因NS的遗传进化情况,本研究采用RT-PCR方法对新疆油鸡分离株的NS基因进行了克隆和序列测定,获得NS蛋白的完整编码序列。将NS基因的核苷酸序列和对应的氨基酸序列与GenBank中已经公布的参考序列进行同源性比较,结果表明,新疆油鸡分离株的NS基因与国内其他物种稳定存在的H9亚型AIV A/Chicken/Beijing/1/1994的NS基因处于同一分支,在系统进化上属于NS基因的A等位基因群。 To evaluate the molecular and evolutional (NS) gene of HgN2 subtype avian influenza viruses characterization of the nonstructural protein isolated from Xinjiang soy sauce chicken,the NS gene of the strain was amplified by RT-PCR,and the complete ORF sequences of NS1 and NS2 protein were obtained. The homology of nucleotide sequences and putative amino acid sequences were compared with several reference strains. The result indicated that NS gene of isolated strain was on the same lineage with A/Chicken/Beijing/1/1994,all of which belong to allele A branch of NS gene.
