突变体黑羽鹌鹑MHC classⅠ第4外显子多态性与新城疫病毒遗传抗性的关联分析 被引量:4

Polymorphismin of exon 4 in MHC class Ⅰ gene and its association with genenic resistance to Newcastle disease virus in black-feather quail

摘要 运用PCR技术扩增突变体黑羽鹌鹑MHC classⅠ基因第4外显子296bp,进行多态性分析,结果显示,该片段有18个突变位点,且均为多态基因座。试验鹌鹑3次感染新城疫病毒F48E9后产生的抗体与18个突变位点进行关联分析,结果表明,第2,4,6,13,14,15突变位点各基因型之间抗体水平差异显著或极显著,这些基因位点可作为抗病力的主要候选基因,为鹌鹑进一步抗病育种提供必要的试验依据。 The exon 4 of the MHC class I gene was amplified with a length of 296 bp,and the pol- ymorphism analysis was performed using PCR technique. The results indicated that there were 18 mutation sites within this fragment,and all of them were located in polymorphic loci. Black feather quail were infected with Newcastle disease virus F48Eg, and the correlation between the induced antibodies and 18 mutation sites was analyzed. The results showed that the antibody level showed significant differences among the phenotypes of mutation sites 2,4,6,13,14 and 15. Hence, these mutation sites can be used as candidate targets for disease resistance. This research provided scien- tific reference for the breeding for disease resistance in quail species.
