响应面优化设计提取葡萄籽中的白藜芦醇 被引量:3

摘要 超声辅助的双水相萃取技术可提高葡萄籽中白藜芦醇的提取率。实验选取"双优红"葡萄酿酒残渣中的葡萄籽为实验材料,优化其中白藜芦醇的提取方法。从16种双水相体系中选出最优体系:聚乙二醇/磷酸氢二钾/水。通过响应面优化实验,得到的最佳提取工艺条件为:超声时间1.5 h,超声温度为50℃,料液比1∶15,乙醇浓度80%。在此条件下,以高效液相色谱法(HPLC)测定"双优红"葡萄籽中白藜芦醇含量为0.38%。 Ultrasonic auxiliary aqueous two-phase extraction technology can improve the extraction rate of resveratrol from grape seeds. With "Shuangyouhong" grape seeds from remains of wine brewing as materials, the optimal extraction method of resveratrol was studied. From 16 kinds of aqueous two-phase system, the optimal system was selected as ethanol/dipotassiium phosphate/water system. With the response surface optimization experiment, the best extraction conditions were as follows: ultrasonic time was 1.5 h, ultrasonic temperature was 50 ℃, solid to liquid was 1 : 15, ethanol concentration was 80%. With this optimum condition, the resveratrol content of the "Shuangyouhong" grape seeds was determined to be 0. 38% by HPLC.
