摘要
目的:探讨乌鳖口服液对自身免疫性卵巢早衰小鼠的影响。方法:通过皮下多点注射小鼠透明带建立自身免疫性卵巢早衰小鼠模型。然后连续4周给予乌鳖口服液进行治疗,每天剂量为(16.5 g/kg)。通过酶联免疫法检测小鼠抗透明带抗体,用免疫组化法测定脾脏T细胞CD_4^+、CD_8^+的含量。同时用免疫组化法测定卵巢Fas、Fas L蛋白的表达。卵巢和脾脏做病理组织学的分析。结果:模型组小鼠抗透明带抗体的浓度(8.74±0.99)U/m L和CD_4^+/CD_8^+比值(1.74±0.31)增加,同时,模型组小鼠卵巢组织的Fas蛋白表达(0.54±0.29)减少,Fas-L蛋白表达(2.25±0.0)增加。与模型组比较,乌鳖口服液组小鼠抗透明带抗体的浓度(7.34±1.21)U/m L明显低于模型组(P<0.05)。脾脏CD_4^+/CD_8^+比值(1.07±0.14)明显低于模型组(P<0.01)。乌鳖口服液组FasL蛋白表达(1.29±0.86)明显低于模型组(P<0.05)。结论:乌鳖口服液具有降低免疫性卵巢早衰小鼠外周血抗透明带抗体浓度的升高,调节CD_4^+/CD_8^+比值和Fas/FasL系统的作用。
Objective: To explore the effects of Wubie oral liquid on autoimmune premature ovarian insufficiency( POI) induced by zona pellucida glycoprotein 3. Methods: BALB/c mice were ntragastrically administered multiple-site subcutaneous injections of mouse zona pellucida glycoprotein 3 and intragastricallyadministered Wubie oral liquid( 16. 5g / kg) for four weeks. To investigate the effects of Wubie oral liquid on BALB / c mice model ofautoimmune premature ovarian insufficiency( POI),anti-zona pellucida antibody( Azp Ab) in peripheral blood of mice were tested by Enzyme-Linked Immuno Sorbent Assay( ELISA),CD4+and CD8+levels in T-cell of mice’s spleens as well as Fas and Fas L protein expression were tested by Immunohistochemistry. The ovarian tissue and spleens histopathological changes were also investigated. Results: The model mice demonstrated increased anti-zona pellucida antibody( Azp Ab) levels in their peripheral blood and CD4+/CD8+in their spleens. Decreased Fasprotein expression and increased Fas L proteinexpression in the ovaries were also observed. Compared with the model group,mice receiving Wubie oral liquid showed significantly decreased concentrationsof Azp Ab and CD4+/CD8+( P 〈 0. 05). Moreover,Wubie oral liquid significantly lowered Fas L protein expression( P 〈 0. 05). After Wubie treatment,the pathological state of the ovary and spleen also improved. Conclusion: The present study reveals that Wubieoral liquid can significantly decrease the concentration of Azp Ab in peripheral blood and regulate the CD4+/CD8+and Fas / Fas L system of mice with autoimmune POI.
