龙胆泻肝汤对变应性接触性皮炎模型小鼠Treg/Th17细胞平衡调节作用的实验研究 被引量:7

摘要 目的:观察变应性接触性皮炎Treg/Th17细胞平衡的变化,探讨龙胆泻肝汤治疗变应性接触性皮炎的部分干预机制。方法:将40只小白鼠随机分为4组:空白组、模型组、龙胆泻肝汤组、氢化可的松琥珀酸钠组,每组10只。采用DNCB皮肤涂抹方法制备小鼠背部ACD模型。模型复制成功后,龙胆泻肝汤组、氢化可的松琥珀酸钠组小鼠均按照成人2倍等效剂量灌胃,连续给药21 d,空白组、模型组给予等量蒸馏水灌胃。实验结束后,取小鼠致敏部位皮肤,对皮肤组织中Foxp3、RORγt、IL-17 mRNA及蛋白的表达水平进行检测。结果:与空白对照组比较,模型对照组、西药组、龙胆泻肝汤组小鼠皮肤组织中Foxp3 mRNA及蛋白表达水平显著降低(P<0.01);而IL-17、RORγt mRNA及蛋白表达水平显著升高(P<0.01)。与模型对照组比较,西药组和龙胆泻肝汤组小鼠皮肤组织中Foxp3 mRNA及蛋白表达水平显著升高(P<0.01);而IL-17、RORγt mRNA及蛋白表达水平显著降低(P<0.01)。结论:Treg/Th17细胞失衡可能是变应性接触性皮炎的发病机制之一,而龙胆泻肝汤可能就是通过作用于该靶点,调节Treg/Th17细胞平衡,而实现其对变应性接触性皮炎的治疗作用。 Objective: To observe Treg / Th17 balance changes of the allergic contact dermatitis in the treatment of Longdan Xiegan Decoction and discuss the intervention mechanism. Methods: Forty mice were randomized into four groups: blank group,model group,Longdan Xiegan Decoction group,hydrocortisone sodium succinate group,10 rats in each group. DNCB was used to establish the mice model of dorsal ACD. When the model was successfully established,Longdan Xiegan Decoction group and hydrogenation cortisone succinate sodium group’s mice were in accordance with 2times of the equivalent dose of adults by gavage,continuous administration for 21 days. The blank group and model group received distilled water by gavage. At the end of the experiment,the sensitized skin was taken out and the expression levels of ROR,t Foxp3,mRNA IL-17 and protein in skin tissue were detected. Results: Compared with the blank control group,model control group,western medicine group and Longdan Xiegan Decoction group’s Foxp3 mRNA and protein expression level decreased significantly( P 〈 0. 01) and IL-17,roryt mRNA and protein expression level increased significantly( P 〈 0. 01). Compared with the model group,western medicine group and Longdan Xiegan Decoction group’s Foxp3 mRNA and protein expression level increased significantly( P 〈 0. 01) and IL-17,roryt mRNA and protein expression level decreased significantly( P 〈 0. 01). Conclusion: Treg / Th17 imbalance may be one cause of allergic contact dermatitis and Longdan Xiegan Decoction may get the therapeutic effect by acting on the target,regulating the balance of Treg / Th17 cells.
