摘要
目的:研究脑泰方提取物(NTFE)对大鼠脑缺血再灌注后内源性神经干细胞增值及Notch1蛋白表达的影响。方法:随机将SD大鼠分为假手术组、模型组及给药组(脑泰方组),以大脑中动脉闭塞(Middle cerebral artery occlusion,MCAO)法建立大鼠局灶性脑缺血再灌注模型,在手术处理后不同时间点给予NTFE药物治疗,利用免疫组织化学方法观察NTFE对脑缺血再灌注后大鼠内源性神经干细胞(NSCs)增殖的影响及Notch1蛋白表达的动态变化。结果:与假手术组比较,模型组Nestin阳性细胞数均增高(P<0.05);各给药组在不同时间点Nestin、Notch1的表达均明显高于同时间点模型组(P<0.05)。结论:NTFE能够促进MCAO模型大鼠缺血再灌注后神经细胞再生,其作用机制可能与Notch信号通路有关。
Objective: To observe effects of Naotaifang on endogenous neural cell regeneration and the expression of Notch1 after cerebral ischemia reperfusion in rats. Methods: SD rats were randomly divided into three groups: sham operation group( A),model group( B) and drug group( NTFE). The local cerebral ischemia model was reproduced by MCAO.The rats were treated with NTFE at different time points after surgery. Use immunohistochemical method to observe proliferation of endogenous neural stem cells( NSCs) and the dynamic change of Notch1 protein. Result: Compared with group A,the Nestin expression was significantly increased in group B( P 〈 0. 05). In drug group at different time points,the expressions of Nestin and Notch1 were significantly increased than that of group B at the same time( P 〈 0. 05). Conclusion: NTFE can promote the regeneration of nerve cells of cerebral ischemia rats after reperfusion and its mechanism may be related to the Notch signal pathway.
