左归丸对骨质疏松症大鼠GPR48、ATF4、BSP表达影响的实验研究 被引量:2

摘要 目的:通过研究摘除卵巢致肾虚骨质疏松症大鼠血清中GPR48、ATF4、BSP的蛋白表达,探讨摘除卵巢致肾虚骨质疏松症的病理机制,并研究左归丸防治疗效及其作用机制。方法:摘除雌性大鼠双侧卵巢的方法复制肾虚骨质疏松症模型,采用具有补肾作用的左归丸对实验大鼠治疗12周,以盖天力片作为阳性对照组,正常大鼠、假手术组作为标准对照组,模型大鼠作为空白对照组,用ELISA法检测肾虚骨质疏松症大鼠血清GPR48、ATF4、BSP的蛋白表达。结果:ELISA方法检测表明:正常大鼠血清中存在GPR48、ATF4、BSP的蛋白表达。模空组大鼠血清中GPR48、ATF4、BSP的蛋白表达水平有明显的下降。左归丸组、盖天力组用药12周后,与模空组相比较,左归丸组可显著上调血清中GPR48、ATF4、BSP的蛋白表达水平。结论:1正常大鼠血清中存在GPR48、ATF4、BSP的蛋白表达;2血清中GPR48、ATF4、BSP的蛋白表达异常变化可能是导致骨质疏松症发生的机理之一;3左归丸能够调控GPR48、ATF4、BSP的蛋白表达,表明左归丸对骨质疏松症有一定的防治作用。 Objective: In order to explore the etiological factors and pathology of the osteoporosis due to kidney deficiency,we studied the expressions of GPR48,ATF4 and BSP in blood serum of the rats with osteoporosis due to kidney deficiency by using ovariectomy. To explore the adjustable function of Zuogui Pill and its regulating mechanism. Methods: By using ovariectomy,we established the rat model of osteoporosis due to kidney deficiency. We treated the osteoporosis rats with Zuogui Pill for 12 weeks. Simultaneously,use Gaitianli tablets,as positive control groups. And rats in the normal group and model group were given no treatment. We used the ELISA method to investigate the expressions of the GPR48,ATF4 and BSP in blood serum of the rats. Result: ELISA detection: GPR48,ATF4 and BSP existed in the normal blood serum. Comparing with the normal group and sham group,the expressions of GPR48,ATF4 and BSP in blood serum of model group decreased evidently. After the precaution treatment with Zuogui Pill and Gaitianli Tablets,he Zuogui Pill group can evidently increase the expressions of GPR48,ATF4 and BSP in blood serum. Conclusion: 1In normal rats,the blood serum can express GPR48,ATF4 and BSP in the protein level. 2The changing of GPR48,ATF4 and BSP protein expressions in the blood serum may be one of the important occurring mechanisms of the osteoporosis due to kidney deficiency. 3The experimental Chinese herbs,which have the function of tonifying kidney,can increase the expressions of the protein of GPR48,ATF4 and BSP in blood serum. It would be preventing and treating the osteoporosis due to kidney deficiency.
