摘要
细胞自噬是细胞代谢的主要途径,广泛参与机体的多种生理病理过程。近年来,随着老年性痴呆又称阿尔兹海默病(Alzheimer’s disease,AD)发病的类淀粉蛋白假说等病理特征以及自噬及其功能联系被相继发现,自噬已成为AD等神经退行性疾病研究的热点。查阅近年国内外文献,将通过归纳阐述自噬与老年性痴呆β-淀粉样蛋白沉积、轴突运输功能障碍、tau蛋白磷酸化以及神经元缺失等神经病理改变相互作用,探讨自噬以及基于自噬治疗的药物在AD发生、发展过程中的作用进行综述、分析、归纳和总结,为AD的治疗和寻找新的临床药物提供新的方向和思路。
Cell autophagy is the main way of cell metabolism and widely involves in a variety of physiological and pathological process. In recent years,as the relation between pathogenesis of Alzheimer’s disease amyloid protein hypothesis and the pathological characteristics of autophagy and its function was found in succession,autophagy has become the research hot spot of neurodegenerative diseases. The author consulted international and domestic literature recently to explore and summarize autophagocytosis and therapeutic drugs based on autophagy in AD’s role in occurrence and development by explaining the autophagy induction and AD beta-amyloid,axonal transport dysfunction,tau protein phosphorylation and neurons lack of nerve pathological changes,providing a new direction and new clinical drugs for AD.
