固本健脑法对老年健忘大鼠模型海马组织中Aβ/RAGE信号通路的影响

摘要 目的:比较固本健脑法、补脾健脑法、补肾健脑法对健忘大鼠海马组织中Aβ/RAGE信号通路的影响,初步揭示固本健脑法改善记忆的部分分子机制。方法:腹腔注射D-半乳糖及Na NO2建立老年健忘大鼠模型,分别灌胃固本健脑方、归脾丸、六味地黄丸和石杉碱甲;并设空白组和模型组,灌以生理盐水。给药结束后,取大鼠海马组织,采用Western blot检测晚期糖化终产物受体(RAGE)、核转录因子p65(NF-κB p65)的表达,ELISA检测β-淀粉样肽1-42(Aβ1-42)、白介素-1β(IL-1β)的含量。结果:与空白组比较,模型组、西药组、补脾组、补肾组上述指标的表达均上调,固本组RAGE、NF-κBp65、IL-1β的表达上调,Aβ1-42则差异无统计学意义。西药组、补脾组、补肾组与模型组比较,上述指标的表达均下调,与固本组比较则上调;补脾组、补肾组、西药组组间比较,差异无统计学意义。结论:固本健脑法、补脾健脑法、补肾健脑法均能在一定程度上改善记忆功能,但固本健脑法作用最佳,固本健脑法改善记忆功能的分子机制与Aβ/RAGE信号通路密切相关。 Objective: To compare the influence of consolidating root and nourishing brain method,strengthening spleen and nourishing brain method and tonifying kidney and nourishing brain method on Aβ / RAGE signaling pathway to introduce the mechanism of consolidating root and nourishing brain method. Method: Intraperitoneal injection of D-alactose and Na NO2 was used to manufacture forgetful old rat models. Gavage animal models with different drugs,ie,prescription on Consolidating Root and Nourishing Brain Recipe,Guipi Pill,Liuwei Dihuang Pill and Huperzine A. After administration,take out the hippocampal area of all groups. Detect the expressions of RAGE and NF-κB p65 by Western blot and test the contents of Aβ1-42 and IL-1β by ELISA. And then compare the differences of the above indices in each groups. Results: Compared with the blank group,model,Western medicine,strengthening spleen and tonifying kidney groups’ indexes were all increased. The consolidating root group’s RAGE,NF-κBp65 and IL-1β were increased and Aβ1-42 had no difference. Compared with the model group,Western medicine,strengthening spleen and tonifying kidney groups’ indexes were all decreased and when compared with the consolidating root group were increased. Comparison among strengthening spleen,tonifying kidney and Western medicine groups had no significance. Conclusion: The results show that all of the four treatment methods can improve the learning and memory function of forgetful old rats to a certain degree,but consolidating root nourishing brain method is obviously the best. And its detail molecular mechanism is closely related to Aβ / RAGE signal pathway. This paper can provide the experimental data and research information for TCM consolidating root and nourishing brain theory and its Chinese patent medicine’s exploitation.
