摘要
目的建立以RP—HPLC法同时测定胆石消胶囊中大黄酸、大黄素、大黄酚和大黄素甲醚的含量测定方法。方法采用Waters SunFire C18柱(250mm×4.6mm,5μm),以甲醇-0.1%磷酸(85:15)为流动相,流速:1.0ml/min,柱温:28℃,检测波长:254am。结果大黄酸、大黄素、大黄酚和大黄素甲醚进样量分别在0.13208~0.79248μg、0.08752~0.52512μg、0.11184~0.67104μg、0.08016~0.48096μg范围内与峰面积呈良好的线性关系,相关系数分别为0.9998、0.9996、0.9994、0.9997,平均加样回收率分别为99.32%、98.80%、99.05%、99.14%,RSD分别为0.76%(n=9)、1.31%(n=9)、1.41%(n=9)、0.94%(n=9)。结论本方法简便易行,专属性强,准确度高,重现性好,可作为胆石消胶囊质量控制方法。
Objective To establish the content determination method of rhein,emodin,chrysophanol and physcion in Danshixiao capsules via RP-HPLC method.Methods Waters SunFire Ciscolumn (250 min×4.6 mm,5 μm) was adopted, and methanol-0.1% phosphate (85:15) was selected as mobile phase,with flow rate:1.0 ml/min,column temperature:28℃, and determination wavelength:254 nm.Results Sample size of rhein,emodin,chrysophanol and physcion was within the range of 0.132 08-0.792 48 μg,0.087 52-0.525 12 μg,0.111 84-0.671 04 μg,and 0.080 16-0.480 96 μg respectively, which showed favorable linear relations with peak areas.The correlation coefficient was 0.9998,0.9996,0.9994 and 0.9997 respectively,the average recovery rate was 99.32%,98.80%,99.05%,99.14% respectively,and RSD was 0.76% (n=9), 1.31% (n=9),1.41% (n =90),0.94% (n =9) respectively.Conclusion The method is easy to operate,with high specificity, high accuracy and good reproducibility,which is able to be taken as the quality control method of Danshixiao capsules.
