摘要 通过对杂交构树的组织培养,研究不同的灭菌方法和激素配比对愈伤组织的诱导、芽的增殖、壮苗及生根的影响。结果表明,最佳灭菌剂为0.5%的Hg Cl2,灭菌15 min后除菌率达到68.2%;诱导愈伤组织分化的最佳培养基为MS+1.5 mg/L 6-BA+1.0 mg/L NAA+0.8%琼脂+3%蔗糖;最佳壮苗培养基为MS+1.5 mg/L 6-BA+0.2 mg/L NAA+0.8%琼脂+3%蔗糖;最佳生根培养基为1/2 MS+0.5 mg/L 6-BA+1.0 mg/L NAA+0.8%琼脂+3%蔗糖;移栽的最佳基质为蛭石,移栽后成活率达到88%。 Based on tissue culture of hybrid Broussonetia papyrifera, this paper studied on the effect of different factors, such as different sterilization treatments and combinations of plant growth regulators. The result showed that the appropriate sterilant agent was0.5% Hg Cl2, the sterilization rate reached 68.2% after 15 min sterilization. The most suitable medium for callus differentiation consisted of MS+1.5 mg/L 6-BA+1.0 mg/L NAA+0.8% agar + 3% sugar; MS+1.5 mg/L 6-BA+0.2 mg/L NAA+0.8% agar+3% sugar was the optimum for strong seedling cultivation; and the best rooting medium was 1/2 MS+0.5 mg/L 6-BA+1.0 mg/L NAA+0.8% ager+3% sugar. The best growth substrate for transplant was vermiculite, the survival rate reached 88%.
