摘要
目的:比较自动加样机联合自动洗板机与手工加样、洗板方式对酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)结果的影响。方法 :采用鸡二脂酰甘油酰基转移酶2(DGAT2)双抗体夹心法试剂盒(供氢体底物为四甲基联苯胺),对倍稀释标准品成5个梯度浓度,采用自动加样机联合自动洗板机和手工加样、洗板,然后经显色后立刻用酶标仪(波长450 nm)检测系列浓度标准品的吸光度(optical density,OD)值,用SPSS软件对浓度和OD值进行相关分析和回归分析。结果:自动加样机联合自动洗板机得出的标准浓度曲线方程的相关系数r、拟合优度R^2及回归系数b值明显高于纯手工加样、洗涤方式,差异有显著性(P<0.05)。结论:在基础医学研究过程应用ELISA需注意不同加样方式和洗涤方式会对ELISA结果造成影响,尽量选用自动化程度较高的方法,以减少误差。
Objective To compare the effect of automatic sample loading and plate washing on ELISA results and that of manual operation. Methods Chicken Diacylgycerol acyltransferase 2(DGAT2) ELISA kit with hydrogen donor substrate as TMB was used to dilute the standard sample to form five groups with gradient concentrations. The samples were treated with automatic loader and plate washer or manual loading and plate washing, and then were measured for optical density(OD) value with an enzyme mark instrument(wavelength 450 nm) immediately after color reaction. Correlation analysis and regression analysis were carried out for the concentration and OD value with SPSS software. Results The correlation coefficient r, goodness of fit R-2, regression coefficients b of automatic mode were significantly higher than those of manual mode(P0.05). Conclusion The mode of sample loading and plate washing influences ELISA results in basic medicine significantly, and automatic mode should be involved as far as possible to decrease errors.
