摘要
目的探讨深圳地区特应性皮炎(atopic dermatitis,AD)患者血清microRNA(miRNA)表达差异,为AD的早期诊断提供生物标志物。方法提取500例AD患者(AD组)和200例正常人(正常对照组)血清总RNA并进行miRNA测序,用实时荧光定量PCR验证差异表达的miRNA。结果与正常对照组相比,AD组患者血清中发现28个具有2倍以上差异的miRNA,其中21个miRNA在外周血血清中含量升高,7个miRNA在外周血血清中含量降低;对28个差异表达的miRNA采用实时荧光定量PCR进行验证,发现hsa-miR-186-5p、hsa-miR-409-3p、hsa-miR-423-5p、hsa-miR-6852-5p表达在AD患者外周血血清中升高。结论hsa-miR-186-5p、hsa-miR-409-3p、hsa-miR-423-5p、hsa-miR-6852-5p在AD的发病过程中可能起关键作用,对AD早期临床诊断有重要意义。
Objective To identify potential diagnostic biomarkers for atopic dermatitis by observing the changes of serum microRNA (miRNA) in atopic dermatitis (AD). Methods Serum miRNAs from 500 patients with AD and 200 healthy controls were sequenced, and the differentially expressed serum miRNAs were validated by fluorescence quantitative real-time PCR technique. Results In serum of AD patients, 28 differently expressed miRNAs (at least a 2-fold change compared with healthy control) were found. There were 21 up-regulated miRNAs and 7 down-regulated miRNAs in AD patients. Quantitative real-time PCR was adopted to confirm the results of sequencing, which showed hsa-miR-186-5p, hsa-miR-409 3p, hsa-miR-423-5p and hsa-miR-6852 5p were up-regulated in peripheral serum samples of AD patients. Conclusion Hsa-miR-186-5p, hsa-miR-409-3p, hsa-miR-423-5p and hsa-miR 6852 5p are the key to the development of atopic dermatitis and play important roles in the early diagnosis.
