摘要
目的观察灯盏花素对H_2O_2致人脐静脉内皮细胞(Human umbilical vein endothelial cells,HUVECs)损伤的保护作用,并探讨其可能的作用机制。方法体外培养稳定传代的HUVECs,以H_2O_2(500μmol·L^(-1))损伤HUVECs 3h建立损伤模型,继而分别采取20,100,200,400mg·L^(-1)浓度的灯盏花素进行干预,观察灯盏花素对H_2O_2损伤的HUVECs细胞保护作用。MTT法检测灯盏花素药物毒性、H_2O_2损伤剂量关系以及细胞活性,试剂盒测定细胞上清液中一氧化(NO)含量、谷胱甘肽-S转移酶(GST)活性,细胞中丙二醛(MDA)水平和超氧化物歧化酶(SOD)活性。结果灯盏花素对HUVECs毒性较小,不同浓度的灯盏花素可显著对抗H_2O_2造成的HUVECs损伤,降低MDA含量,增强SOD活性,促进NO释放,升高GST活性。结论灯盏花素对H_2O_2所致的HUVECs损伤具有明显的保护作用,其机制可能与抑制损伤细胞的脂质过氧化,提高机体抗氧化酶的活性,促进一氧化氮(NO)的合成有关。
Objective To observe the protective effects of Breviscapine on hydrogen peroxide impaired HUVECs and to explore its possible mechanism. Methods The HUVECs was modeled by 500μmol·L^(- 1) H_2O_2 injured for 3h and incubated by breviscapine( 20,100,200,400 mg·L^(- 1)). The drug toxicity of breviscapine,dosage relations of hydrogen peroxide impair and cells survival rate were evaluated by MTT method. The content of nitric oxide( NO) and the activity of glutathione- s- transferase( GST) in cell culture supernates,the content of malondialdehyde( MDA) and the activity of superoxide dismutase( SOD) in cells were examined by kits. Results Breviscapine produced a less toxicity to HUVECs. The different doses of breviscapine could effectively reduce MDA content,increase no content,SOD and GST activity and significantly improve the HUVECs injured by H_2O_2. Conclusion Breviscapine was showed a significantly protective effects on HUVECs injured by H_2O_2. Its mechanism might relate to inhibit lipid peroxidation,increase antioxidase activity of body and facilitate synthesis of NO.
