摘要
目的考察ZTC天然澄清剂联合大孔树脂对白子草总多糖的纯化效果,确定白子草总多糖的纯化工艺。方法以絮体形态、溶液状态、多糖转移率为指标,考察加样顺序、药液浓度、药液p H、絮凝温度、絮凝剂用量等因素对ZTC总多糖纯化效果的影响;以多糖转移率为指标,考察大孔树脂型号、药液p H、药液浓度、吸附流速、药材树脂(干)比、树脂柱径高比、洗脱液p H、洗脱流速等因素对大孔树脂总多糖纯化效果的影响。结果建立了ZTC纯化白子草总多糖的工艺,白子草总多糖纯度从26.34%提高至48.86%;建立了大孔树脂纯化白子草总多糖的工艺,白子草总多糖纯度从48.86%提高至65.26%。结论本研究采用ZTC天然澄清剂联合大孔树脂技术建立了白子草总多糖的纯化工艺,为白子草降血糖活性成分的开发及进一步利用奠定了基础。
Objective The purpose of this study was to establish the total polysaccharides purification technology of Gynurae Divaricata by studying the purification effect of ZTC natural clarifying agent combined with macroporous resin on the total polysaccharides from Gynurae divaricata. Methods The flocculation state,solution clarity,and transferring ratio of total polysaccharides were used as indicators to investigate the influence of adding order of clarifying agent,concentration of extracting solution,p H value of extracting solution,flocculation temperature,clarify agent dosage on the total polysaccharides purification efficiency. The transferring ratio of total polysaccharides was used as indicator to investigate the influence of macroporous resin model,p H value of extracting solution and elution agent,concentration of extracting solution,flow speed of adsorption,ratio of polyamide to crude drug,ratio of diameter to height of polyamide column,and flow speed of the elution agent on the total polysaccharides purification efficiency. Results The total polysaccharides purification technology by ZTC was established with content of total polysaccharides increased from 26. 34% to 48. 86%. The total polysaccharides purification technology by macroporous resin was established with content of total polysaccharides increased from 48. 86% to 65. 26%. Conclusion The total polysaccharides purification technology of Gynurae divaricata was established by ZTC natural clarifying agent combined with macroporous resin in this study,which laid a foundation for further using and development of active components with blood sugar reducing activity in Gynurae Divaricata.
