穿心莲组培苗的壮苗生根与移栽基质研究 被引量:3

摘要 目的探讨不同因子对穿心莲组培苗壮苗生根的影响,并考察生根苗的移栽基质,为完善穿心莲种苗的离体快繁体系提供试验依据。方法以穿心莲的组培苗为实验材料,分别考察了大量元素的用量、α-萘乙酸(NAA)与蔗糖的含量、接种时组培苗的叶片数等因子对组培苗壮苗生根的影响,考察不同基质对生根苗移栽后生长的影响。结果以1/2 MS培养基中为基础培养基、培养基中含0.1 mg/L的NAA与20 g/L的蔗糖、接种含6片叶的组培苗等条件有利于组培苗的壮苗生根,以河沙与蛭石1∶1为基质有利于生根苗移栽后的生长。结论大量元素的用量、NAA与蔗糖的含量、接种时组培苗的叶片数等因子对组培苗的壮苗生根具有显著的影响,蛭石与河沙组成的基质更适合生根苗的移栽与生长。 Objective To study the factors of strengthening and rooting of tissue culture seedlings and transplanting matrix of Andrographis paniculata,for the purpose of completing its in vitro rapid propagation system. Methods Tissue culture seedlings were selected as experiment material. The factor,such as the amount of macroelement,1- naphthaleneacetic acid( NAA) and sucrose,and the leaf number of tissue culture seedlings when inoculate,which were examined impact on the strengthening and rooting of them. The transplanting matrix were also studied after transplanting the seedlings. Results The amount of macroelement in1 / 2 MS culture medium,which contains 20 g / L sucrose and 0. 1 mg / L NAA,and inoculated the seedlings contain six leaves were suitable for strengthening and rooting of tissue culture seedlings. The matrix consisted with sand and vermiculite at the ratio of 1:1 was suitable for transplanting. Conclusion The factors mentioned above had significant effect on strengthening and rooting of tissue culture seedlings. The consisted of sand and vermiculite was suitable for rooted seedlings transplant.
