摘要
目的以皮质酮肾阳虚动物模型为研究对象,采用正交设计研究多个针刺参数,探索影响针刺效应的主次因素和针刺参数的最佳组合。方法采用L9(34)正交表对4个参数(穴位、捻针角度、频率、时间)3水平安排试验,运用计算机控制针刺手法仪进行干预,通过ELISA检测血清中T_4、TRH的水平和RT-PCR检测垂体组织中TSH mRNA的表达来比较上述4因素的主次。结果对针刺效应的影响由大到小依次为穴位、捻针角度、捻针频率、捻针时间,其中穴位是影响针刺效应的主要因素(P<0.01);针刺捻针的角度、频率、时间都是影响针刺效应的次要因素(P均>0.05)。结论针刺治疗肾阳虚大鼠的最佳针刺参数为针刺肾俞穴,捻针角度为90°,频率为120次/min,捻针时间60 s。该研究中穴位选择对疗效的影响最大,其次为捻针角度,再次是捻针频率,影响最小的为捻针时间。
Objective To explore primary- secondary factors that influences acupuncture effectiveness and the best combination of acupuncture parameters. Orthogonal design was adopted to study several factors that influence acupuncture effectiveness,rats model of kidney- YANG deficiency made by hydrocortisone as the study objects. Methods 4 influential factors( i. e. acupoints,needle- twisting angle,needle- twisting frequency and manipulation time) plus 3 levels of each factor were arranged to L9( 34)orthogonal table. Treat kidney- YANG deficiency rats by computer controlled acupuncture manipulation device. Concentration of serum T_4 and TRH were determined by ELISA method and mRNA expression of pituitary TSH by RT- PCR to illustrates the primary- secondary factors that could affect the acupuncture effectiveness. Results Among 4 factors mentioned above,the order of influential factors from major to small was: acupoints,needle- twisting angle,needle- twisting frequency and manipulation time,among which,acupoints was the major factors in acupuncture effectiveness( P〈0. 01); needle- twisting angle,needle- twisting frequency and manipulation time all were secondary role in acupuncture effectiveness( P〉0. 05 and P〉0. 05). Conclusion 1The best acupuncture parameters for treating kidney- YANG deficiency rats were: Shenshu acupoint( B23),90°of needle- twisting angle,120 tpm of needle- twisting frequency and 60 s of manipulation time; 2In this study,acupoints became the biggest influential factor,the next was needle- twisting angle,the secondary was needle- twisting frequency,and the last one was manipulation time.
