摘要
目的:研究益艾康胶囊对奈韦拉平在大鼠体内药动学的影响,为益艾康胶囊在临床合理用药提供依据。方法:实验大鼠分别灌胃给予奈韦拉平和奈韦拉平+益艾康胶囊,并通过LC/MS/MS测定2种给药方式后血浆中奈韦拉平浓度,随后使用Phoenix Win Nonlin 6.1软件计算药动学参数。结果:符合方法学考察要求:血药浓度在5~500 ng·mL^-1范围内线性关系良好,日内和日间精密度均〈10.0%,提取回收率89.8%~107.0%,稳定性较好,受基质影响较小。由2组药动学主要参数得知MRT0~t和MRT0~∞均显著升高(P〈0.05),Cmax显著降低(P〈0.05),其他参数无统计学意义。结论:益艾康胶囊确实对奈韦拉平在大鼠体内药动学产生了影响。
Objective: To study the effect of Yiaikang( YAK) capsules on the pharmacokinetics of nevirapine( NVP) in rats and provide the basis for its rational use in clinic. Methods: HPLC-MS / MS was used to measure the NVP concentration in the plasma of rats after intragastric administration of NVP and NVP + YAK,respectively. Then the pharmacokinetic parameters were calculated by Phoenix Win Nonlin 6. 1. Results: The HPLC-MS / MS method met the requirements of the methodological well. The calibration curve for plasma NVP had good linearity in the concentration range of 5 ~ 500 ng·mL^-1. The intar-day and inter-day RSDs were less than10. 0%,abd the extraction and recovery rates ranged from 89. 8% to 107. 0%. The data indicated that the stability of the samples was good and there was little substrate effect. The main pharmacokinetic parameters of NVP,MRT 0 ~ t and MRT 0 ~ ∞,were significantly increased in the NVP + YAK group( P〈0. 05),and Cmaxwas significantly decreased( P〈0. 05). However,the difference in the other pharmacokinetic parameters showed no statisticalsignificance. Conclusion: Yiaikang capsules have significant effect on the pharmacokinetics of nevirapine in rats.
