摘要
以草莓主栽品种鬼怒甘的茎尖为材料,研究了Hg Cl2浓度及其消毒时间、基础培养基、白砂糖、6-苄氨基腺嘌呤(6-BA)、萘乙酸(NAA)、吲哚丁酸(IBA)、吲哚乙酸(IAA)、活性炭(AC)等9种主要因素对草莓茎尖组织培养的影响,分析筛选出了一套能够产出具有优良素质草莓种苗的组织培养技术,建立起了比较完善的草莓茎尖组织培养快繁体系。结果表明:不同因素对草莓茎尖组织培养的影响较大,尤其是影响草莓的形态发育(分化、增殖、生根和生长)。最佳外植体消毒方法是0.20%Hg Cl2 8-11min,适合初代培养的培养基为MS+6-BA 0.5-1.0mg/L+NAA 0.01mg/L+白砂糖30g/L,增殖继代培养基为MS+6-BA 0.5-1.0mg/L+NAA 0.01-0.05mg/L+白砂糖20-30g/L,生根培养基为MS+NAA 0-0.01mg/L+IBA 0-0.1mg/L+IAA 0-0.1mg/L+白砂糖30g/L。
Taking one main strawberry variety of Guinugan as sample,experiments were carried out to study the effects of 9 factors(Hg Cl2,basal medium,white granulated sugar,etc.) on tip-shoot tissue culture of strawberry,and then to screen out a tip-shoot tissue culture technology for strawberry,which could produce better quality seedlings,and eventually to establish a relatively complete tip-shoot tissue culture and rapid propagation system in strawberry. The results showed that 9 factors had great effects on strawberry tip-shoot tissue culture.The best sterilization approach for explants was to treat 8-11 min by 0.20% Hg Cl2,the suitable primary culture medium was MS+6-BA 0.5-1.0mg/L+NAA 0.01mg/L+white granulated sugar 30g/L,the suitable proliferation medium was MS+6-BA 0.5-1.0mg/L+NAA 0.01-0.05mg/L+white granulated sugar 20-30g/L,and the suitable rooting medium was MS+NAA 0-0.01mg/L+IBA 0-0.1mg/L+IAA 0-0.1mg/L+ white granulated sugar 30g/L.
