摘要
目的:摸索制备黄芪甲苷纳米乳的最佳处方。方法:伪三元相图法分析不同处方比例对微乳区形成面积的影响。考察PC/Tween 80、Km、AS/O值、含水量对纳米乳载药量的影响。综合考虑伪三元相图及载药量分析结果,确定制备黄芪甲苷纳米乳的最佳处方比例。结果:当PC/Tween 80=3:7、2:8,Km=3:1、2:1时所形成的微乳区面积较大。当含水量为60%,PC/Tween 80=2:8,Km=2:1,AS/O=9:1时纳米乳载药量最大。综合考虑,最终确定黄芪甲苷纳米乳的最佳处方比例为PC:Tween 80:无水乙醇:IPM:H2O(w/w)=4.8%:19.2%:12%:4%:60%。结论:综合考虑伪三元相图及载药量分析结果,确定纳米乳最佳处方更具全面性。
Object To explore the best composition for prescription of Astragaloside Ⅳ nanoemulsion. Methods Pseudo-ternary phase diagrams was used to analyze the impact of the composition of nanoemulsion on microemulsion area. The impact of value of PC/Tween 80, Km, AS/O and moisture on drug loading were studied. The results of both Pseudo-ternary phase diagrams and RP-HPLC were taken to consideration to explore the best composition for prescription of Astragaloside Ⅳ nanoemulsio. ResultsMicroemulsion area were larger when PC/Tween 80 was 3:7 or 2:8, Km was 3:1 or 2:1. Drug loading were higher with composition of 60% moisture, PC/Tween 80 as 2:8, Km as 2:1 and AS/O as 9:1. After considering, we finaly set PC: Tween 80: cosurfactant: IPM: H2O(w/w)as 4.8%: 19.2%: 12%: 4%: 60% to be the best composition for prescription of Astragaloside Ⅳ nanoemulsio. Concludes Taking the results of both Pseudo-ternary phase diagrams and drug loading to consideration is comprehensive for exploration of the best nanoemulsion composition.
