摘要
let-7通过影响靶基因的表达水平而参与细胞的生长、分化、凋亡等。为了研究let-7-5p在鸭上的序列特征、表达模式及其对鸭骨骼肌发育的可能作用,本文应用本课题组前期mi RNA测序数据,通过生物信息分析技术和分子生物学技术,研究了鸭let-7-5p序列特征、表达模式、其靶基因及靶基因的功能注释。结果如下:1鸭let-7-5p成熟序列是TGAGGTAGTAGGTTGTATAGTT;2鸭let-7-5p在北京鸭孵化第11天(E11)至E27胸肌内的表达量逐渐增加。组织间表达模式显示,在鸭E27心脏内的表达量最高(3.33),在肝脏、皮脂、肌胃、小肠和腹脂的相对表达量较低;3鸭let-7-5p靶基因的GO条目中许多条目与骨骼肌发育密切相关。研究结果能为进一步研究let-7-5p对鸭骨胳肌发育的作用奠定基础。
Let-7 often involves in the growth,differentiation, apoptosis of cell through repressing the expression of its targets. To investigate the sequence characteristics,expression pattern,and the potential effect of let-7-5p on skeletal muscle development of duck, we carried out a further analysis based on our previous miRNA-seq data by using bioinformaties and molecular biology technologies. The results were listed as follow. (1)The mature sequence of duck let-7-5p is TGAGGTAGTAGGTI'GTATAGT]'. (2)The expression levels of let-7-5p in Pekin duck breast muscle increased stably from Ell to E27. And the expression level in heart was the highest among the different tissues at E27 stage, the expression levels in liver, skin fat,muscular stomach, small intestine and abdominal fat were lower than that in other tissues. (3) The GO results of let-7-Sp targets showed that many GO terms involved in skeletal muscle development. In conclusion,the results of this paper could provide a basis for the further investigation of the effects of let-7-Sp on duck skeletal muscle developmeut.
