摘要
[目的]观察滋阴抑亢调冲汤对卵巢过度刺激综合征(ovarian hyperstimulation syndrome,OHSS)模型大鼠肿瘤坏死因子α(tumor necrosis factoralpha,TNF-α)、血清雌二醇(estradiol,E_2)含量及腹腔毛细血管通透性的影响,探讨滋阴抑亢调冲汤治疗OHSS的疗效及其作用机制。[方法]22日龄雌性Wistar大鼠60只,随机分为正常对照组、模型对照组、滋阴抑亢调冲汤组和吲哚美辛组,每组15只,除正常对照组外,其余3组均按照改良的Ujioka方法皮下注射孕马血清促性腺激素(pregnant mare serum gonadotropin,PMSG)和人绒毛膜促性腺激素(human chorionic gonadotropin,HCG)建立OHSS模型,造模成功后分别灌胃给予相应剂量的药物,用药1周后处死大鼠,肉眼、光镜观察卵巢组织形态学改变,检测腹腔冲洗液中亚甲蓝含量,采用放射免疫法检测大鼠血清E_2水平,ELISA法检测血清TNF-α含量,免疫组化法检测卵巢组织TNF-α蛋白的表达。[结果]滋阴抑亢调冲汤能降低OHSS模型大鼠血清E_2、TNF-α及腹腔冲洗液中亚甲蓝含量,差异有统计学意义(P<0.05,P<0.01),下调卵巢组织TNF-α蛋白的表达,差异有统计学意义(P<0.05),改善卵巢组织形态学变化。[结论]滋阴抑亢调冲汤可通过降低血清E_2、TNF-α含量,下调卵巢组织TNF-α蛋白的表达,改善腹腔毛细血管通透性,对OHSS发挥治疗作用。
[Objective] To observe the effects of Ziyinyikangtiaochong decoction on tumor necrosis factor-alpha(TNF-α), the content of serum estradiol and vascular permeability of peritoneal in ovarian hyperstimulation syndrome(OHSS) model rats so as to explore efficacy and mechanism of the decoction.[Methods] Sixty female Wistar mice of 22 days were randomly divided into 4 groups(normal control group, model group, Ziyinyikangtiaochong decoction group, indomethacin group), with 15 mice in each group. Except for normal control group, the other three groups were inclined to use pregnant mare serum gonadotropin and human chorionic gonadotropin to establish the ovarian hyperstimulation syndrome(OHSS) model by improved Ujioka methods. Each group was given appropriate doses of various drug by gavage. After one week, make anesthesia to the rats in each group, remove both ovaries blooded, observe ovarian tissue morphology with eye and light microscopy,the methylene blue content of peritoneal fluid was measured, radioimmunoassay was used to detect content of serum E2 and ELISA method was used to detect content of tumor necrosis factor-α(TNF-α), immunohistochemistry was used to detect the protein expression of TNF-α in ovarian tissue for the rats. [Results] Ziyinyikangtiaochong decoction can reduce the content of serum E2 and TNF-α,the methylene blue content of peritoneal fluid(P〈0.01), down-regulate the protein expression of TNF-α in ovarian tissue(P〈0.05) and improve the morphological changes of ovary in OHSS rats. [Conclusion] Ziyinyikangtiaochong decoction can treat OHSS by correlating with reducing abdominal capillary permeability, content of serum E2 and TNF-α, the protein expression of TNF-α in ovarian tissue.
