摘要
目的:通过观察苏木提取物对心脏移植模型大鼠心肌病理形态学及生长因子表达的影响,探讨苏木提取物对慢性排斥反应的作用机理,为从中药学中研发出新型的免疫抑制剂奠定工作基础。方法:应用改良Ono术式行大鼠腹腔心脏移植,结合Cs A灌胃建立心脏移植慢性排斥动物模型。选择移植心脏跳动良好的大鼠随机分为3组。苏木组予苏木乙酸乙酯提取物溶液灌胃;环孢素组予Cs A溶液灌胃;模型对照组按体质量计算予等体积橄榄油灌胃,连续灌胃7 d后取下移植心脏。观察移植术后受体的一般情况,移植心脏收缩力情况,移植心脏心肌病理形态学改变,并进行慢性排斥反应诊断分级;采用Real-time-PCR法检测各组大鼠移植心脏心肌生长因子PDGF-A、VEGF-C、FGF-1、FGF-2的基因表达水平。结果:与模型对照组相比,苏木组和环孢素A组PDGF-A的表达水平降低,差异有统计学意义(P<0.05);苏木组和环孢素A组移植心脏心肌组织PDGF-A的表达水平相当,差异无统计学意义(P>0.05);与模型对照组相比,苏木组和环孢素A组VEGF-C的表达水平降低,差异有统计学意义(P<0.05);苏木组和环孢素A组移植心脏心肌组织VEGF-C的表达水平相当,差异无统计学意义(P>0.05);与模型对照组相比,苏木组和环孢素A组FGF-1的表达水平降低,差异有统计学意义(P<0.05);与苏木组相比,环孢素A组FGF-1的表达水平降低,差异有统计学意义(P<0.05)。与模型对照组相比,苏木组和环孢素A组FGF-2的表达水平降低,差异有统计学意义(P<0.05);与苏木组相比,环孢素A组FGF-2的表达水平降低,差异有统计学意义(P<0.05)。结论:苏木乙酸乙酯提取物能降低大鼠慢性排斥模型移植心肌PDGF-A、VEGF-C的基因表达,作用与Cs A相当。苏木乙酸乙酯提取物能降低大鼠慢性排斥模型移植心肌FGF-1、FGF-2的基因表达,作用不及Cs A。
Objective:To observe the effect of ethyl acetate extracts from sappanwood on the pathomorphology of transplanted heart and the expression of growth factor with chronic rejection after allogenic cardiac transplantation of rats, and to preliminarily discuss the mechanism of ethyl acetate extracts of sappanwood for resisting the chronic rejection, laying a foundation for the research of new immunosuppressants. Methods:The allogenic cardiac transplantation model was estab-lished by using the improved Ono operation skill and the CsA. The rats with successful operations were divided into three groups. The rats in sappanwood group were given olive oil containing sappanwood. The rats in experimental group were given olive oil containing CsA. The rats in control group were given olive oil. Observe their general conditions, the survival of the transplanted hearts and the heartbeats. Observe the recipients' general conditions and the heartbeats. Observe the pathological changes of transplanted hearts with light microscope according to the degree of chronic rejection. Then detect the expression levels of PDGF - A, VEGF - C, FGF - 1 and FGF - 2 in rats' myocardium of every group by using the method of Real - time - PCR. Results : Compared with the model control group, PDGF - A was expressed less in Sap- panwood group and CsA group, and there was a significant difference (P 〈 0.05 ). Compared with the model control group, VEGF, C was expressed less in Sappanwood group and CsA group, and there was a significant difference ( P 〈 0. 05 ). Compared with the model control group, FGF - 1 was expressed less in Sappanwood group and CsA group, and there was a significant difference(P 〈 0.05 ) ;compared with the Sappanwood group, FGF- 1 was expressed less in CsA group, and there was a significant difference ( P 〈 0.05 ). Compared with the model control group, FGF - 2 was ex- pressed less in Sappanwood group and CsA group,and there was a significant difference(P 〈0.05) ; compared with the sion:The ethyl acetate extracts from sappanwood can reduce the expressions of PDGF - A and VEGF - Cin myocardial cells in transplanted hearts and the effect is similar to CsA. The ethyl acetate extracts from sappanwood can reduce the expressions of FGF - 1 and FGF,- 2 in myocardial cells in transplanted hearts and the effect is inferior to CsA.
