摘要
在CTAB法的基础上,以榛树休眠芽为材料,使用组织研磨棒快速研磨代替液氮研磨,使用极低毒性的PVP(聚乙烯吡咯烷酮)代替高毒的β-巯基乙醇,建立了适于榛树基因组DNA快新方法。经检测验证,该方法具有快速、低毒、准确的特点。
Based on the CTAB method,taking ormant buds of Corylus heterophylla as materials,grind rapidly by using tissue grinding rod in place of liquid-nitrogen grinding.highly toxic β-mercaptoethanol was replaced by using extremely low toxicity of PVP(polyvinylpyrrolidone);new method which is optimal for fast-extracting DNA of genome of Corylus heterophylla was established.Result shows that the method is fast,low toxicity and accurate by verification.
