高效液相色谱法测定感冒软胶囊中对乙酰氨基酚、琥珀酸多西拉敏、氢溴酸右美沙芬的含量

Content Determination of Acetaminophen,Doxylamine Succinate,Dextromethorphan Hydrobromide in Soft Capsule by HPLC

目的：建立高效液相色谱法测定感冒软胶囊中对乙酰氨基酚、琥珀酸多西拉敏、氢溴酸右美沙芬含量的方法。方法：色谱柱为Agilent Zorbax C_（18）（4.6 mm×150 mm,5μm）,流动相A为乙腈,流动相B为缓冲液（pH=3.0）,梯度洗脱,流速为1.2 ml/min,检测波长为275 nm,柱温为35℃,进样体积为20μl。结果：对乙酰氨基酚、琥珀酸多西拉敏、氢溴酸右美沙芬的线性范围分别为2.62~26.23μg/ml（r=0.999 9）、2.51~25.14μg/ml（r=0.999 8）、5.68~56.78μg/ml（r=0.999 9）;回收率分别为100.6%、101.1%、100.2%,RSD分别为0.4%、1.2%、0.4%（n=9）。结论：本方法操作简便快捷,结果准确可靠,可用于感冒软胶囊中对乙酰氨基酚、琥珀酸多西拉敏、氢溴酸右美沙芬的含量测定。

OBJECTIVE： To establish a method for the content determination of acetaminophen,doxylamine succinate,dextromethorphan hydrobromide in Soft capsule by HPLC. METHODS： Separation was carried out on a Agilent Zorbax C_（18）（ 4. 6 mm × 150 mm,5 μm） column with mobile phase A of acetonitrile,mobile phase B of buffer solution（ pH = 3. 0） in gradient elution at a flow rate of 1. 2 ml / min,the detection wavelength was 275 nm,the column temperature was 35 ℃,and the injection volume was 20 μl. RESULTS： The linearity range of acetaminophen,doxylamine succinate,dextromethorphan hydrobromide were 2. 62-26. 23 μg / ml（ r = 0. 999 9）,2. 51-25. 14 μg / ml（ r = 0. 999 8） and 5. 68-56. 78 μg /ml（ r = 0. 999 9）,respectively. The average recovery rates were 100. 6%,101. 1% and 100. 2%,with RSD of 0. 4%,1. 2% and 0. 4%（ n = 9）,respectively. CONCLUSIONS： The method is simple,rapid,and accurate in the content determination of acetaminophen,doxylamine succinate,dextromethorphan hydrobromide in Soft capsule.
