摘要
目的探讨子痫前期孕妇血清及胎盘组织中IFN-γ诱导蛋白10(IP-10)及其受体CXCR3的表达情况。方法选择同期住院的晚期正常妊娠、轻度子痫前期和重度子痫前期孕妇各20例。采用ELISA、实时荧光定量RT-PCR和Western blot等方法检测血清IP-10水平、CXCR3 m RNA表达水平,以及胎盘组织中IP-10、CXCR3蛋白和m RNA表达水平;并对子痫前期孕妇IP-10与CXCR3进行Pe a rs on相关分析。结果与正常妊娠组比较,轻、重度子痫前期孕妇血清IP-10水平和CXCR3 m RNA表达水平均有不同程度的增高(均P<0.05),且均为重度子痫前期组高于轻度子痫前期组(均P<0.05)。此外,轻、重度子痫前期孕妇胎盘组织中IP-10、CXCR3蛋白和m RNA水平均明显高于正常妊娠组(均P<0.05)。Pearson相关分析显示,子痫前期孕妇血清IP-10水平与外周血CXCR3 m RNA表达水平呈正相关(r=0.914,P<0.05);子痫前期孕妇胎盘组织中IP-10 m RNA表达水平与CXCR3m RNA表达水平呈正相关(r=0.907,P<0.05)。结论 IP-10及其受体CXCR3表达上调参与子痫前期病程,子痫前期孕妇血清、胎盘组织中IP-10与CXCR3均呈正相关,可能与子痫前期的发病有关。
Objective To investigate the expression of IFN- γ inducible protein 10（IP- 10） and its receptor CXCR3 in placenta and serum of gravidas with preeclampsia.Methods Samples of placenta and serum were collected from normal gravidas,gravidas with mild preeclampsia and gravidas with severe preeclampsia with 20 cases in each group.The protein and mRNA expression of IP- 10 and CXCR3 in placenta and serum was detected by ELISA,real-time PCR and Western blot,respectively.Results ELISA showed that serum IP- 10 levels in patients with severe or mild preeclampisa were significantly higher than those in normal pregnancy group（P〈0.05）.The mRNA expression of CXCR3 in peripheral blood of patients with severe or mild preeclampisa was significantly higher than that in normal pregnancy group（P〈0.05）.Real- time PCR and Western blot showed that the expression levels of IP- 10 and CXCR3 mRNA and protein in the placenta of severe or mild preeclampisa groups were significantly higher than those in normal pregnancy group（P〈0.05）.Conclusion The elevated expressions of IP- 10 and its receptor CXCR3 in the placenta and serum may be involved in the pathogenesis of preeclampsia.
