碳光子治疗仪局部照射对大鼠腓肠肌闭合性急性损伤的影响 被引量:3

Effects of local irradiation with carbon photon therapeutic apparatus on acute gastrocnemius muscle injury in rats

摘要 目的观察碳光子治疗仪局部照射对大鼠腓肠肌闭合性损伤后急性炎症反应、肌细胞损伤及肌肉再生情况的影响.方法:70 只9 周龄雄性SD 大鼠随机分为3 组:空白对照组(n=5)、损伤组(n=35) 及损伤后照光组(n=30).以500 g 砝码从50 cm 高度自由坠落击打大鼠腓肠肌,制备肌肉损伤模型.制模成功后损伤后照光组受损部位使用YW-0828A 型碳光子治疗仪( 波长320-5 000 nm,功率密度11.5 mW/cm2) 每天光照20 min,连续10 d;损伤组及空白对照组同-部位接受相同机器的假性照射.在损伤后0( 损伤后即刻)、1、2、3、5、7、14 d 测量大鼠后肢损伤处的周径;取血进行中性粒细胞计数,磷酸肌酸底物法检测血清肌酸激酶,ELISA 法检测血清C 反应蛋白(CRP) 水平;取肌肉组织进行组织病理学观察.结果:大鼠后肢在击打后即刻明显肿胀,伤后0、1、2、3、5、7 d 较对照组后肢腓肠肌周径明显增加(P 〈 0.01).损伤后照光组在损伤后第1、2、5、7 天后肢腓肠肌周径较损伤组明显减小,损伤后照光组于伤后7、14 d,损伤组于伤后14 d 达到空白对照组水平.损伤后照光组在损伤后1、3、5 天中性粒细胞计数,损伤后1、2、3、5、7 天的血清肌酸激酶含量及损伤后1、2、3、5 天的血清CRP 水平均明显减少(P 〈 0.01).损伤组腓肠肌于损伤后即刻出现出血、水肿、变性及部分腓肠肌断裂,第7 天可见新生的肌纤维,水肿减轻,第14 天肌肉可见纤维化;损伤后照光组第7 天镜下可见排列整齐的肌纤维和较少的成纤维细胞,第14 天可见新生肌纤维排列整齐.结论:碳光子治疗仪照射可促进炎症吸收、减轻炎症反应,减少肌细胞损伤,促进骨骼肌的恢复. Objective: To observe the effects of carbon photon instrument therapy on acute inflammatory response,muscle cell damage and muscle regeneration following contusion of gastrocnemius muscle in rats. Methods: Seventy maleSD rats aged 9 weeks old were randomly divided into 3 groups: control group (n=5), contusion group (n=35), and postcontusionirradiation (PCI) group (n=30). The gastrocnemius damage model was prepared by free fall of 500 g weight fromthe 50 cm height. In PCI group, irradiation with YW-0828A type carbon photon instrument (wavelength 320-5 000 nm,power density 11.5 mW/cm2) was administered to the contused gastrocnemius muscle for 10 days with 20 minutes each day.The same parts of rats received sham irradiation with the same instrument in the contusion group and control group. Muscleinjurymodel was evaluated on 0 (immediately), 1, 2, 3, 5, 7, 14 days after injury by measuring the circumference at theinjury site of the hind limb of rat. At the same time, blood was sampled, and neutrophils were counted, the levels of creatinekinase and C reactive protein (CRP) were determined by phosphocreatine substrate method and ELISA respectively, and thepathological changes in the gastrocnemius muscle were observed. Results: The hind limbs of the rats were obviously swollenimmediately after injury. Circumference of hind limb in contusion group was significantly larger than that of the controlgroup on 0,1,2,3,5,7 days after injury (P〈0.01). Irradiation significantly decreased the circumference of the hind limb, thedifference was significant between the PCI group and contusion group at 1,2,5,7 days after contusion. The circumference ofthe hind limb was similar to that of the control group at 7 and 14 days in the PCI group or at 14 days in the contusion group.After irradiation, the neutrophils counts on 1,3,5 days, the levels of creatine kinase on 1,2,3,5,7 days and CRP on 1,2,3,5days after injury was significantly reduced compared to the contusion group. After injury, the gastrocnemius in the contrusion group developed the hemorrhage, edema and degeneration of the gastrocnemius and fracture of a part of gastrocneminusimmediately after the injury. newly formed muscle fibers and alleviated edema were seen on 7 days , while muscle fibrosiswas seen on the14th day. In PCI group, well-arranged muscle fibers and less fibroblasts were seen on the 7th day, and visiblenew muscle fibers arranged regularly on the 14th day. Conclusions: Irradiation with carbon photon instrument can promotethe absorption of inflammation, reduce the inflammatory reaction, modify the damage to muscle cells, and promote therecovery of skeletal muscle.
