摘要 目的研究胃癌相关miR-193b调控胃癌细胞增殖的功能作用,并通过研究其与Akt信号通路的关系探讨miR-193b的调控机制。方法在PTEN基因特异性敲除(PTEN^(-/-))小鼠胃癌组织中,Northern Blot法检测miR-193b的表达变化;在胃上皮正常细胞系GES-1和5种胃癌细胞系AGS、BGC-823、SGC-7901、N87、SNU-16中,Real-Time PCR和Western Blot分别检测miR-193b与p Akt、Akt的表达水平;激活或抑制胃癌细胞Akt信号通路,RealTime PCR检测miR-193b表达改变;在胃癌细胞BGC-823、SGC-7901中过表达miR-193b,检测细胞的增殖变化。结果 miR-193b在Akt异常激活的小鼠胃癌组织中表达显著下调;胃癌细胞中Akt信号通路激活程度与miR-193b表达水平呈负相关,Akt信号通路抑制miR-193b表达;过表达miR-193b抑制胃癌细胞增殖。结论 miR-193b抑制胃癌细胞增殖并受Akt信号通路负向调节,其表达下调可能参与Akt信号通路调控的细胞增殖与胃癌发生。 Objective To investigate the regulation role of miR - 193b on proliferation of gastric cancer cells,and explore regulating mechanism of miR - 193b by analysing correlation between miR - 193b expression and Akt signaling pathway activation. Methods The expres-sion of miR - 193b in gastric cancer tissues of PTEN knockout mouse was checked by Northern Blot;The expression of miR - 193b in 5 gastric cancer cell lines and 1 epithelial cell line was detected by Real - Time PCR,as well as the protein level of pAkt and Akt were checked by Western Blot in the same cell lines;The regulatory role of miR - 193b on gastric cancer cells proliferation were explored with cell counting. Results miR- 193b was down - regulated in gastric cancer tissues of mice with aberrant activation of Akt pathway for PTEN knockout. There was a negative correlation between miR - 193b level and Akt pathway activity in gastric cell lines,and Akt pathway inhibited the expression of miR - 193b in gas-tric cancer cells. Over - expression of miR - 193b significantly suppressed the proliferation of gastric cancer cells. Conclusion miR - 193b ex-pression is negatively regulated by Akt signaling pathway,and might be involved in the proliferation and tumorigenesis regulated by Akt signaling pathway.
