缺营养状况下培美曲塞对小鼠肺癌细胞系LLC化疗的影响

摘要 目的观察小鼠肺癌细胞系LLC分别在全营养和缺营养(无氨基酸无血清培养)情况下,使用化疗药物培美曲塞的效果。方法噻唑蓝(MTT)方法检测培美曲塞对小鼠肺癌细胞LLC细胞的半抑制浓度(IC50),决定培美曲塞干预浓度。将小鼠肺癌细胞系(LLC)培养在全营养和无氨基酸无血清培养基中,分别用培美曲塞处理12 h,用MTT法检测细胞活性,用采用凯基活细胞/凋亡细胞/坏死细胞鉴别试剂盒(AO/EB荧光显微镜法)检测细胞凋亡。结果培美曲塞对LLC的IC50为28.67 mM。将0、3.58 mM、7.16 mM、14.33 mM、28.67 mM和57.34 mM做为培美曲塞的干预浓度,培美曲塞浓度在以上干预浓度下,无营养培养基中(NF),对LLC的抑制率分别为0,17.4%,26.6%,37.7%,39.0%,45.8%;全营养组(FN)中的抑制率为0,23.7%,30.2%,48.9%,57.4%,67.4%。缺营养的状况下,培美曲塞对小鼠肺癌细胞LLC的杀伤作用明显下降(P<0.05)。缺营养和全营养的情况下,LLC细胞的凋亡率分别为24.9%,22.6%;缺营养情况下和全营养情况下,肺癌细胞在化疗药物作用下的死亡率分别为25.3%,34.2%。结论缺营养的情况下,小鼠肺癌细胞对化疗药物培美曲塞的敏感性降低。 Objective To observe the effect of pemetrexed on LLC lung cancer cell line mouse lack of nutrition group and full nutrition group. Methods MTT method was used to detect the half inhibitory concentration(IC50)of pemetrexed on LLC cells. Pemetrexed treated mouse lung cancer cell line(LLC)cultured in FN and NF for 12 h,respectively. MTT was used to detect cell activity,KGI living cells/ cell apoptosis /necrosis cell identification Kit(AO/ EB fluorescence microscopy method)was used to detect the apoptosis. Results IC50 of pemetrexed on LLC was 28. 67 mM. According to the IC50 value,the intervention concentration of pemetrexed was choosed as 0,3. 58 mM,7. 16 mM,14. 33 mM, 28. 67 mM,57. 34 mM. On these different concentrations of pemetrexed,the inhibitory rates of LLC in NF were 0,17. 4% ,26. 6,% 37. 7% , 39. 0% ,45. 8% ,while the inhibition rate in FN was 0,23. 7% ,30. 2% ,48. 9% ,57. 4% ,67. 4% ,respectively. In NF,LLC lung cancer cellsˊs survive decreased significantly( P ﹤ 0. 05). In NF and FN,the apoptosis rate of LLC cells were 24. 9% ,22. 6% ,respectively;In NF and FN treated with pemetrexed,the mortality of lung cancer cells was 25. 3% ,34. 2% ,respectively. Conclusion In the lack of nutrition condi-tions,the sensitivity of mouse lung cancer cells to chemotherapeutic drugs pemetrexed were reduced.
