摘要
目的:研究沙棘总黄酮( total flavones of hippophae )对东莨菪碱( scopolamine )所致记忆障碍小鼠的作用及其与海马内乙酰胆碱酯酶(AChE)浓度的关系。方法将昆明种小鼠随机分为4组(n=10):生理盐水(NS)组、东莨菪碱2 mg· kg ^-1(Scop)组、沙棘总黄酮100 mg· kg^-1(Tfh100)组、沙棘总黄酮200 mg· kg^-1(Tfh200)组。在避暗实验中,小鼠分别腹腔注射东莨菪碱建立记忆障碍模型,观察沙棘总黄酮对记忆障碍小鼠潜伏期和错误次数的影响。避暗实验结束后立即断头取脑,分别检测每组小鼠海马中AChE的浓度。结果与NS组相比,Scop组的错误次数明显增多,潜伏期明显缩短,AChE的浓度显著升高(P均<0.01);与Scop组相比,Tfh100、Tfh200组潜伏期明显延长,错误次数明显减少,AChE的浓度显著下降(P均<0.01)。结论沙棘总黄酮对东莨菪碱所致小鼠记忆障碍有保护作用,其机制可能与其降低海马内AChE的浓度有关。
Objective To explore the effects of total flavones of hippophae on scopolamine induced memory impair -ment in mice and the its relationship with the concentration of acetylcholinesterase ( AChE) in the hippocampus .Meth-ods Mice were randomly divided into four groups (n =10): a normal saline (NS) group, a 2 mg/kg scopolamine (Scop) group, a 100 mg/kg total flavones of hippophae (Tfh100) group and a 200 mg/kg total flavones of hippophae (Tfh200) group.A mouse model of memory impairment was established through intraperitoneal injection of scopolamine . Then, the effects of total flavones of hippophae on the latency and number of errors were observed in the step -through test.Immediately after the step-through test, the brains were removed to detect the concentration of AChE in the hippo-campus of mice .Results Compared with the NS group , the Scop group presented remarkably higher number of errors , shortened latency, and increased concentrations of AChE (P〈0.01).Compared with the Scop group, the Tfh100 and Tfh200 groups showed markedly extended latency , less number of errors , and decreased concentrations of AChE ( P〈0.01).Conclusion Total flavones of hippophae have protective effects on scopolamine -induced memory impairment in mice, which may be related to decreased concentration of AChE in the hippocampus .
