摘要
目的:研究毛兰素对人结肠癌Caco-2细胞增殖的抑制作用及其诱导细胞凋亡的分子机制。方法:采用SRB法检测毛兰素对Caco-2细胞增殖的抑制作用,用Hoechst33342染色法观察细胞的形态学改变,流式细胞术检测细胞的凋亡率和细胞周期;蛋白免疫印迹法(Western Blot)检测细胞凋亡相关蛋白Caspase-3的表达水平。结果:与对照相比,毛兰素能抑制结肠癌细胞Caco-2的增殖,且抑制率随着药物浓度与时间增加,呈剂量时间效应,48 h半数抑制浓度IC50为0.845μg/m L;毛兰素能诱导Caco-2细胞凋亡,并诱导细胞周期阻滞于G_2期;Caspase-3活性裂解片段表达增高。结论:毛兰素对肿瘤细胞的增殖具有一定抑制作用且通过线粒体途径诱导Caco-2细胞凋亡。
Objective: Growth inhibition effects of erianin on human colorectal cell line Caco-2 cells and its possible mechanism were studied.Methods: SRB assay was employed to evaluate the anti- proliferation effect of erianin on Caco-2 cells.The morphological changes of treated Caco- 2 cells were observed with 33342 staining.Cell cycle distribution and the proportion of apoptotic cells were evaluated using flow cytometry.Western Blotting analysis was used to detect the expressions of apoptosis related protein Caspase- 3. Results: Erianin inhibited proliferation of Caco-2 cells in a dose- and time- dependent manner and leaded to cell apoptosis.The IC50 value was 0.845 μg / m L after 48 h exposure to erianin. Cell cycle was arrested in G_2 phase. It was also observed that the expression of activity fragmentation of Caspase-3 showed a significant increase.Conclusion: Erianin could inhibit the proliferation of Caco- 2 cells and the apoptosis of Caco- 2 cells induced by erianin may be associated with mitochondrial pathway.
